Ethyl caprate (≥99%) was purchased from Shanghai Macklin Biochemical Technology Co., Ltd. (Shanghai, China), and n-alkane mixed standard C7-C40 was purchased from o2si (o2si smart solutions—an LGC Standards Company, Charleston, SC, USA).

Comprehensive Phytochemical Profiling of T. hemsleyanums

HPLC-grade acetonitrile and formic acid were purchased from Thermo Fisher Scientific Company (USA). Ultra-pure water was purchased from Watsons Company (China). All other reagents were of analytical grade.
The reference standards such as kaempferol, isoquercetin, astragalin, rutin, quercetin, baicalein, and palmitic acid were purchased from Herbpurify Co., Ltd. (Chengdu, China); emodin-8-O-β-D-glucoside, quercitrin, vitexin, and vitexin-rhamnoside were purchased from Yuanye Bio-Technology Co., Ltd. (Shanghai, China); isorhamnetin, catechins and epicatechin were purchased from Institutes for Food and Drug control (Beijing, China).
The plant materials of T. hemsleyanums were collected from different provinces in China, as Zhejiang (ZJ1, ZJ2, and ZJ3), Guangxi (GX1, GX2, and GX3), Yunnan (YN1, YN2, and YN3), Fujian (FJ1, FJ2, and FJ3), Guizhou (GZ1, GZ2, and GZ3), and Hubei (HB1, HB2, and HB3). The botanical authentication was performed by Professor Lingchuan Xu from Department of Pharmacognosy, Shandong University of Traditional Chinese Medicine. A voucher specimen (no. TH20171201~TH20181118) was deposited in Key Lab for Natural Product, Shandong Province. The dried rhizome parts of T. hemsleyanums were used for further analysis.

Extraction and Characterization of Bioactive Compounds

Chemical reagents like ethanol, methanol, acetonitrile, and formic acid were all of chromatographic grade from Thermo Fisher Scientific Inc. (Waltham, MA, USA). Using a Milli-Q Plus instrument from Merk KGaA, 18.2 MΩ.cm water was made (Darmstadt, Germany1). Folin–Ciocalteu and aluminum nitrate were supplied by Solarbio Science & Technology Co., Ltd. (Beijing, China). Chemical standards including chlorogenic acid, ferulic acid, vitexin, rutin, gallic acid, myricitrin, morin, glycitein, wogonin, kaempferol-3-O-sophoroside, liquiritigenin, and butin were obtained from Yuanye Biological Technology Co., Ltd. (Shanghai, China), with >98% purity. The extract was concentrated on Bond Elut PPL 6 cc, 500 mg C18 solid phase extraction (SPE) cartridge that was purchased from Agilent Technology Co., Ltd. (Beijing, China). High-glucose (4.5 g/L) Dulbecco’s modified eagle’s medium (DMEM), which contains 10% (v/v) heat-inactivated fetal bovine serum, 100 µg/mL of streptomycin, and 100 U/mL of penicillin, was obtained from Gibco Laboratories (Carlsbad, CA, USA). Escherichia coli 0111: B4 LPS was a Sigma product (St. Louis, MO, USA).

(Shanghai, China). Thermo Fisher Scientific (Waltham, MA, USA) provided the methanol (HPLC quality), formic acid (99.9%, LC-MS grade), and acetonitrile (LC-MS grade). Cellulase (T richoderma Vride G, 400 u/mg), hemicellulase (Aspergillus niger, 20 u/mg), and pectinase (Aspergillus niger, 500 u/mg) were obtained from Yuanye Bio-Technology Co., Ltd. (Shanghai, China).

Comprehensive Analysis of Litsea gracile Herb

Chromatographic‐grade acetonitrile and methanol were obtained from Fisher Scientific. Formic acid in MS grade was provided by Sigma‐Aldrich. Fifteen batches of Lgracile herbal materials were collected from their main origin in China and identified by Prof. Jiyu Gong of Changchun University of Chinese Medicine, School of Pharmaceutical Sciences. The information of samples is shown in Table S1.
