Plasma Renin and Inflammatory Markers Measurement

Blood was collected by tail bleed for serial measurements and finally by terminal bleed. The plasma fraction was separated by centrifugation upon collection and stored at -80°C until assay. Renin activity in plasma was assessed via production of angiotensin I from angiotensinogen using a commercially available GammaCoat Plasma Renin Activity 125I RIA kit (DiaSorin, Stillwater, MN), using porcine angiotensinogen (A2283, Sigma-Aldrich, St. Louis, MO) substrate. Urine albumin and creatinine were measured on spot urine sample using Albuwell and Creatinine kit (Exocell, Philadelphia, PA). Commercially available ELISA kits were used for the measurements of serum CCL2 and IL-6 (R&D Systems, Minneapolis, MN).

Urinary Biomarkers of Renin-Angiotensin

Check if the same lab product or an alternative is used in the 5 most similar protocols
Urinary concentration of AGT and sPRR were measured by ELISA kits (IMMUNO-BIOLOGICAL LABORATORIES IBL, Co, Fujioka, Japan, Cat. # IB08003 and 27782, respectively). Ang II concentrations in urine were determined by radioimmunoassay (RIA), as previously described26 (link). Results are expressed in fmol/24 h of urine. Urinary renin content (URC) was determined by using modified protocols from the PRA assay [GammaCoat Plasma Renin Activity 125I RIA kit (DIASORIN, Stillwater, MN, USA)]. Urine samples were spiked with 1 μM synthetic renin substrate tetradecapeptide (RST, Sigma). The generated Ang I was determined by RIA. To exclude the effect of peptidases, identical urine samples-RST with the specific renin inhibitor WFML peptide (ANASPEC; Fremont, CA) were used as controls.

Quantifying Renin in Cell Cultures

Check if the same lab product or an alternative is used in the 5 most similar protocols
Renin content in cell culture media was determined by using modified protocols from PRA assay (GammaCoat Plasma Renin Activity 125I RIA kit [DiaSorin, Stillwater, MN]) as previously described (Gonzalez et al. 2015, 2016). Data were expressed by ng Ang I formed per hour per mL of cell culture media.

Metabolic and Hormonal Biomarkers in Fasting Individuals

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood and urine samples were taken after about 12 h of fasting. Plasma C-reactive protein (CRP), sodium, potassium, glucose, creatinine, uric acid, triglyceride, and total, high-density (HDL) and low-density lipoprotein (LDL) cholesterol concentrations were determined using Cobas Integra 800 (F. Hoffmann-LaRoche Ltd, Basel, Switzerland). Insulin and parathyroid hormone (PTH) were determined using electrochemiluminescence immunoassay (Cobas e411, Roche Diagnostics). Estimated glomerular filtration rate (eGFR) was calculated using the CKD-EPI cystatin C equation [22 (link)], and insulin sensitivity was evaluated by the quantitative insulin sensitivity check index (QUICKI) [23 (link)]. Plasma renin activity (GammaCoat® Plasma Renin Activity 125-I RIA Kit, DiaSorin, Saluggia, Italy) and aldosterone concentration (Active® Aldosterone RIA, Beckman Coulter, Fullerton, CA, USA) were determined using commercial kits. Urine albumin excretion was determined using immunoturbidimetry.
The normal reference range for uric acid in Finland is 230–480 µmol/l for men, 155–350 µmol/l for women aged 18–49 years, and 155–400 µmol/l for women aged ≥ 50 years [24 ].

Estimated glomerular filtration rate (eGFR) was calculated using the Chronic Kidney Disease – Epidemiology collaboration (CKD-EPI) cystatin C equation27 (link). Insulin sensitivity was evaluated by the quantitative insulin sensitivity check index (QUICKI)28 (link), and homeostatic model assessment of insulin resistance (HOMA-IR)29 (link). In addition, the subjects were invited to participate in a standard 75-g oral glucose tolerance test (OGTT) for the determination of the Matsuda index30 (link).

Electrolytes were measured by selective ion method, albumin by the bromocresol green colorimetric method, total proteins by kinetic assay, creatinine by the Jaffeḿ ethod, urea by kinetic-UV assay and glucose levels by using the enzymatic method with a hexokinase. All these parameters were determined using an Automatic Analizator Roche-Hitachi, Cobas c311. Plasma renin activity was measured by Radioimmunoassay, using a Gamma Coat Plasma Renin Activity 125 I RIA Kit, DiaSorin.

Urine and blood samples were taken after 12 h of fasting. Measurements of plasma sodium, potassium, calcium, C-reactive protein (CRP), cystatin C, glucose, creatinine, uric acid, triglycerides, and total, high-density and lowdensity lipoprotein cholesterol concentrations were determined using Cobas Integra 700/800 (F. Hoffmann-La Roche Ltd, Basel; Switzerland) or Cobas6000, module c501, and plasma insulin with an electrochemiluminescence immunoassay on Cobas e411 analyzer (F. Hoffmann-La Roche Ltd). Blood cell count was calculated by ADVIA 120 or 2120 (Bayer Healthcare, Tarrytown, New York, USA). Radioimmunoassays were used for the analyses of plasma renin activity (GammaCoat Plasma Renin Activity 125-I RIA Kit; DiaSorin, Saluggia, Italy) and aldosterone concentration (Active Aldosterone RIA, Beckman Coulter, Fullerton, California, USA). Glomerular filtration rate was evaluated using CKD-EPI-creatinine-cystatin-C formula [21] , and insulin sensitivity using the quantitative insulin sensitivity check index (QUICKI) [22] .

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
