CFTR Transfection in Human Hepatocytes

Human hepatocytes (HL-7702) were obtained from the Japanese Collection of Research Bioresources and cultured in RPMI-1640 medium (Thermo, USA) containing 10% fetal bovine serum (FBS) (Hyclone, USA) and 1% penicillin (Thermo, Waltham, MA, USA) humidified with 5% CO2 air at 37 °C. The cells were transfected with Ad-CFTR when they were 80% confluent. The control cells were transfected with Ad-GFP. After 48 h, the transfection efficiency was detected by fluorescence microscope, and then the cells were treated with L-Hcy (100 μmol/L) (Sigma, USA) for another 24 h for further experiments.

Cell Line Culture Protocols

The cells were cultured in Dulbecco’s Modified Eagle Medium (DMEM) (Gibco BRL, Grand Island, NY, USA) containing 10% fetal bovine serum (FBS) (Gibco) and incubated at 37°C in a humidified chamber containing 5% CO2.

Homocysteine Exposure in Hepatocytes

Hepatocyte cell line HL‐7702 was obtained from the Japanese Collection of Research Bioresources. The cells were cultured in 1640 plus 10% serum and 1% antibiotics. When the cells were 70% confluent, serum was deprived for 24 h to synchronize cells, cells were then cultured with 5% fetal calf serum before treatment with 0, 50, 100, 200, and 500 μM Hcy (Sigma‐Aldrich). To compensate for the short half‐life of Hcy, the compound was replenished every 12 h.
