Sperm medium

Manufactured by Cook Medical
Sourced in Australia

Sperm Medium is a lab equipment product designed to support the survival and motility of sperm cells during assisted reproductive procedures. It provides a balanced, isotonic solution to maintain the optimal environment for sperm during handling and processing.

Sperm Preparation Using Density Gradient

Check if the same lab product or an alternative is used in the 5 most similar protocols
Sperm samples were prepared using density gradient technique according to manufacturer’s protocol (Sperm Gradient kit, COOK medical, Australia). Briefly, density gradient media was prepared by layering the 40% medium over the 80% medium, and two ml of the liquefied semen sample were added over the upper layer. After centrifugation (380 g for 10 minutes), most of the supernatant was gently removed and the pellet was resuspended in two ml of Sperm Medium (Sperm Medium, COOK medical, Australia) to remove the density gradient medium. At the end of the centrifugation (670 g for 5 minutes), the final pellet was resuspended in the Sperm Medium.

Sperm Motility Enrichment via Swim-up

Check if the same lab product or an alternative is used in the 5 most similar protocols
From each semen sample, one aliquot was used to prepare the motile sperm—enriched fraction using the swim-up technique [49 (link)]. Briefly, one volume of semen was placed in a tube and was overlaid by one volume of culture medium (Sperm Medium, COOK medical, Australia) according to the manufacturer’s instructions. The tube was incubated for 45 min at 37°C under 5% CO2, 5% O2 and 90% N2. The supernatant was aspirated and transferred to an empty tube: replicate measurements of sperm concentration, sperm motility, and sperm morphology were performed according to the World Health Organization criteria 2010 [48 ] (Fig 1).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
