Transgenic Mouse Models for Immune Research

The following mice on a C57BL/6 background were purchased from The Jackson Laboratory and used at 10 weeks of age unless otherwise stated: Cxcl12DsRed knockin (stock no. 022458) (23 (link)), which were used in isolation or crossed with the following: Tcrα-/- (stock no. 002116, (24 (link)), Rag2-/-(stock no. 008449) (25 (link)), Foxn1Cre (stock no. 018448)(26 (link)) and Rosa26-stop-EYFP (stock no. 006148) (27 (link)). Control mice for experiments involving and Tcra-/- and Rag2-/- mice were heterozygous littermate controls. RANKVenus BAC transgenic mice were generated as described previously (28 (link)). Husbandry, housing, and experimental methods involving mice were performed at the Biomedical Services Unit at the University of Birmingham in accordance with the local Ethical Review Panel and UK Home Office Regulations (Animal project Licence number P3ACFED06).

Conditional Knockout Mice for Lamin Research

All mouse experiments were approved by the Institutional Animal Care and Use Committee of Carnegie Institution for Science. Lmnb1f/f allele was derived from Lmnb1tm1a(EUCOMM)Wtsi by the EUCOMM project. The conditional Lmnb2 allele, Lmnb2f/f, was derived from Lmnb2tm1a(KOMP)Wtsi (KOMP project) by breeding with ACTB‐FLPe mice to remove neomycin cassette flanked by Frt sites. Lmnaf/f and Lmnb1/ alleles were generated in house by Dr. Youngjo Kim and were reported previously (Kim & Zheng, 2013). All mice were further backcrossed to the C57BL/6 background for at least 6 generations. LckCre, Foxn1Cre, K5CreETT2, K8CreERT2, and OT‐II mice were purchased from the Jackson Laboratory. K5CreETT2 and K8CreERT2 mice were further backcrossed to the C57BL/6 background for at least 2 generations. 20‐mon‐old female C57BL/6 mice were purchased from National Institution on Aging. Other mice in cohort experiments at the indicated ages were raised by the mouse facility of Carnegie Institution for Science. All animals were housed under a 12‐/12‐hr light–dark cycle and fed ad libitum.
