Comprehensive Immunohistochemical Profiling

IHC-Based HER2 Expression Scoring

HER2 expression was examined in the tumor tissue by IHC19 (link),49 (link). Briefly, 4-μm sections were deparaffinized, rehydrated, and treated with 3% hydrogen peroxide-methanol at room temperature for 5 min and then heated in a water bath at 98 °C for 40 min in the target retrieval solution (pH 9.0; Dako, Glostrup, Denmark). The sections were blocked with 8% skim milk in Tris-buffered saline (TBS) at 37 °C for 40 min and then incubated with a rabbit polyclonal anti-human c-erbB-2 oncoprotein (HER2/neu, 1:100 dilution; Dako) at 37 °C for 40 min. The sections were incubated with the EnVision polymer reagent for rabbit IgG (Dako) at 37 °C for 60 min. Then, the reaction products were visualized using 3,3-diaminobenzidine. HER2 immunoreactivity was quantified based on a previous study19 (link). Samples with HER2 IHC scores of 0 and 1 were classified as negative, and those with scores of 2 and 3 were classified as positive.

Immunohistochemical Analysis of CK5, CK20, and HER2
