Culturing Human Lung Cancer Cell Lines

The human non-small cell lung cancer cell lines A549 (Adenocarcinoma) and NCI-H460 (large cell carcinoma) were purchased from the National Centre for Cell Science (NCCS), Pune, India. The cell lines were cultured using a complete medium which includes, DMEM, 10% FBS, and 1% antibiotic antimycotic solution (penicillin, streptomycin, and amphotericin B). The cultured cell lines were incubated in a CO2 incubator at an atmospheric temperature of 37°C and supplied with 5% CO2.

Cell Line Cultivation Protocol

Human non-small cell lung cancer cell lines (A549 and NCI-H460) and human monocytic THP-1 cell lines were procured from the National Centre for Cell Science (NCCS) Pune, India. Cells were cultured in RPMI1640 media supplemented with 10% FBS and 1% Penicillin-Streptomycin at 37°C in a humidified chamber with 5% CO2. MRC-5 human lung fibroblast cells were obtained from the ATCC, USA; and BEAS-2B human bronchial epithelial cells was a generous gift from Prof. Anita K. Verma. MRC-5 and BEAS-2B were cultured in EMEM and LHC-9 media, respectively, supplemented with 10% FBS and 1% Penicillin-Streptomycin solution at 37°C in a humidified chamber with 5% CO2.

Culturing NSCLC Cell Lines A549 and NCI-H460

The human NSCLC cell lines A549 and NCI-H460 were procured from the National Centre for Cell Science (NCCS), Pune, Maharashtra, India. Both the cell lines were maintained in RPMI-1640 medium supplemented with 10% FBS, 1% 200 U/mL of penicillin, 1% 0.2 mg/ml of streptomycin, and 1% 200mM L-glutamine in a humidified, 5% CO2, 37˚C incubator (Eppendorf, Germany). The media was changed every 2-3 days, and the cells were separated via trypsinization using 0.25% trypsin/0.038% EDTA when they reached 70-80 % confluence.
