Ficoll paque separation media

Manufactured by GE Healthcare

Ficoll-Paque separation media is a density gradient medium used for the separation and isolation of different cell types, such as mononuclear cells, from whole blood or other biological samples. It functions by allowing the various components of the sample to separate based on their unique densities when centrifuged.

Measuring IFN-γ production in T cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood from 3 healthy donors was used to separate peripheral blood mononuclear cells, using Ficoll-Paque separation media (GE Healthcare). MLR was essentially performed as previously described.38 (link) Further details are in the supplemental Material and methods. After 48 hours, all samples were treated for 5 hours with Golgi plug protein transport inhibitor, brefeldin (BD Biosciences) to inhibit cytokine accumulation in the Golgi complex and processed to measure interferon-γ (IFN-γ) production by CD4+ and CD8+ T cells by flow cytometry.

Isolation of Leukemia and Normal Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Primary leukemia specimens (n = 3) were obtained from ALL patients upon ethical approval from Weill Cornell Medicine Institutional Review Board (IRB; # 0909010629, #0910010677). De-identified normal peripheral blood (NPB) samples (n = 3) were purchased from the New York Blood Center (New York, NY). Mononuclear cells (MNCs) were isolated by density separation using the Ficoll-Paque separation media (GE Healthcare, #17-1440-03), as manufacturer instructions, and cryo-stored in liquid nitrogen until use.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
