Biodegradable PLA filament for drug-loaded hydrogels

Polylactic acid (PLA) was chosen as biodegradable thermoplastic printer filament for FDM (melting point 220ºC, density 1.25 g/cm 3 ), supplied by Leon3D (Spain). Hydroxypropyl methylcellulose METHOCEL™ K4M (Colorcon Ltd., UK) was used as hydrophilic polymer to prepare drug-loaded hydrogels for IVF. Anhydrous theophylline (Acofarma, Spain) has been used as a model drug. Eudragit FS30D (Evonik, Germany) was chosen as delaying release polymer.

Glipizide HPMC Polymer Formulations

Formulation and Characterization of Ibuprofen Delivery

Ibuprofen (IBU) was purchased from Farmasino Pharmaceuticals Co. Ltd. (Jiangsu, China). Pharmaceutical grade hydroxypropyl methylcellulose (METHOCEL K4M) was kindly donated by COLORCON (Dartford, Uk). Hydroxypropyl methylcellulose (Pharmacoat 603, Hypromellose 2910) and hypromellose acetate succinate (AQOAT-AS-LMP) were donated by Shin-Etsu (Tokyo, Japan) respectively. Tween 80 was purchased from Sigma-Aldrich (Gillingham, UK). The HPLC solvents were analytical grade and purchased from Fisher Chemicals (UK).

Metformin Quasi-Emulsion Tablet Formulation

Metformin hydrochloride (MF, Auro Laboratories Ltd., Mumbai, India) was dissolved in demineralized water and crystallized in technical-grade acetone (density used for calculations: 0.784 g/mL at 25 °C [14 (link)]). Two types of hypromellose (HPMC, Pharmacoat® 603, Shin-Etsu Chemical Co., Ltd., Tokyo, Japan, and Methocel™ K4M, Colorcon, Dartford, UK) were used as polymeric stabilizers of the quasi-emulsion.
For tableting, hyprolose (HPC SSL SFP, Nippon Soda, Tokyo, Japan) was used as a dry binder, and magnesium stearate (Parteck® LUB MST, Merck KGaA, Darmstadt, Germany) as a lubricant.
Caffeine (BASF, Ludwigshafen, Germany) was used as a tracer, to determine the residence time distribution.

Controlled Release Loxoprofen Sodium Formulation

Hydrochloric acid (37% w/v), potassium dihydrogen phosphate (≥99.5% w/w), sodium hydroxide (≥99% w/w), acetonitrile (≥99.99% w/w), and acetone (≥99.8% w/w) were all purchased from Merck (Darmstadt, Germany). Entire chemicals and solvents used in the study were of analytical grade.
