BE(2)-C cells transfected with pEGFP-N1/pEGFP-ZNF32 were cross-linked with 1% formaldehyde for 15 min at room temperature. Formaldehyde was quenched with 125 mM Glycine for 5 min, and the cells were then collected and washed. Cells and cell nuclei were lysed sequentially. The extract was sonicated and incubated overnight with anti-GFP antibody (ab183734, Abcam, United Kindom) and anti-IgG (ab171870, Abcam, United Kindom) at 4°C. The bound DNA was analyzed using PCR.
