Collagen-Induced Arthritis Model in Mice

A mixed emulsion of bovine collagen type II (2 mg/ml, Chondrex, Redmond, WA, USA) and complete Freund’s adjuvant (CFA, 2 mg/ml, Chondrex) or incomplete Freund’s adjuvant (IFA, Chondrex) were prepared according to a standard CIA protocol by Chondrex, Inc. Then, 100 μl of the CFA emulsion was subcutaneously immunized to DBA/1 mice (male, 6-weeks-old) at the base of the tail, followed 3 weeks later by immunization with 100 μl IFA emulsion. scR2agoTNF-Fc and saline (negative control) were administered i.p. twice a week from day 22 after immunization.

Induction and Assessment of Adjuvant-Induced Arthritis in Rats

Male Sprague-Dawley rats (160–180 g) were obtained from Liaoning Changsheng Biotechnology Co., Ltd. All animal experimental protocols were approved by the Institutional Animal Care and Use Committee of Jilin University (Number of permit: SY202111027). Animal care was following the Laboratory Animals-Guidelines for ethical review of animal welfare in China (GB/T 35892–2018). All the rats were fed with a chlorophyll-free chow before the experiment.
Complete Freund adjuvant (CFA) containing 10 mg/mL of heat-killed Mycobacterium tuberculosis was obtained from Chondrex, Inc. According to the protocol for AIA rat model, 50 μL of CFA was subcutaneously injected into the left footpads of the rats. On day 4 after CFA injection, severe and acute inflammation of the joints was observed, and continued to develop thereafter. On day 14 after CFA induction, AIA rat model was successfully induced. During this process, the severity of arthritis in rats was assessed every two days by clinical scores of the joint swelling severity and thickness of the hind paws. The arthritis severity of the rats was scored as follows: 0 = no erythema or swelling, 1 = slight swelling and confined erythema, 2 = slight swelling and extended erythema, 3 = moderate swelling and extended erythema, 4 = severe swelling and widespread erythema.

Rheumatoid Arthritis Induction in Minipigs

Rheumatoid arthritis was induced according to a previous study [19 (link)] with minor modifications. Briefly, a total of 15 specific pathogen free (SPF) minipigs (3 to 4 months old males with body weight of 15–20 kg) were obtained from OPTIPHARM (OPTIPHARM Co., Ltd., Korea). All animals were sedated with an intramuscular injection of ketamine (20 mg/kg) and xylazine (2.5 mg/kg). Anesthesia was performed by isoflurane inhalation. For all animals, left knee joint cavities were directly administered with a mixture of 0.5 mg/kg bovine type II collagen (CII; Chondrex, WA, USA) emulsified with an equivalent volume of 2 mg/ml CFA (Chondrex) intra-articularly on Day 1. The second immunization was performed on Day 22 using the same procedure except that IFA (Chondrex) instead of CFA was used.

Knee Injection and Tracing Protocol

All knee injections were conducted under anesthesia (100 mg/kg ketamine and 10 mg/kg xylazine, intra-peritoneally) through the patellar tendon. For experiments with retrograde tracing, 1.5 µl of fast blue (FB) or titer-matched AAV-PHP.S viruses were injected intraarticularly into knee joints (~4x10 11 vg of AAV-PHP.S-CAG-dTomato and ~5x10 11 vg of AAV-PHP.S packaged with Gi and Gq-DREADD into each knee). The titer values were chosen based on a pilot experiment conducted with AAV-PHP.S-CAG-YFP, injection of 2 ul (~3x10 11 vg) and 10 ul (~10 ^12 vg) labelled a similar number of neurons as assessed by acute culture. For experiments involving inflammation, 7.5 µl CFA (10 mg/ml, Chondrex) was injected into the left knee and Vernier's calipers were used to measure knee width (as before (7) ) pre-and 24hour post-CFA injection.

Immunization and Th17 Cell Analysis

Collagen-Induced Arthritis Model in Mice

CIA was induced as previously described32 (link). Briefly, 9-week-old sex-matched WT (C57BL6/J) and Eaf2−/− mice (generated on a pure C57BL6/J background) were injected intravenously with 100 μl of a 1/1 (v/v) emulsion of 0.05 M acetic acid containing 100 μg of chick type II collagen (20012, Chondrex) and CFA (7023, Chondrex). The day of the first immunization was defined as day 0. Twenty days after the primary immunization, the mice were boosted with 100 μg of collagen II in IFA via the same route. The severity of arthritis was evaluated every 3 days after the boost by visual inspection. Each paw was scored for clinical signs of arthritis as described52 . The clinical score for each mouse was the sum of the four paw scores, which results in a maximum score of 16. On the final day of the experiments, all mice were anaesthetized and their blood was collected by cardiac puncture.

Intra-TMJ Injection of CFA Model

Collagen-Induced Arthritis Model in Mice

C57BL/6 mice aged 8– 12 weeks were immunized with the emulsion of complete Freund's adjuvant (CFA, catalog number: 7023, Chondrex, Redmond, WA) and 100 μg of chicken type II collagen (catalog number: 20012, Chondrex) in a 1:1 mixture (total 100 μl) subcutaneously into the base of the tail on day 018 (link). The booster immunization of CII with incomplete Freund's adjuvant (catalog number: 7002, Chondrex) was performed on day 21. Recombinant human G-CSF (Filgrastim, Neupogen, Amgen, Inc., Thousand Oaks, CA) was injected at 10 μg peritoneally once daily from day 20 to day 23. Vehicle (water) or 50 mg/kg of JBI-589 was orally administered twice daily from day 20 to day 56. The severity of arthritis was evaluated using the following clinical scoring system for each limb: 0, normal; 1, swelling in one finger joint; 2, swelling in more than one finger joint or wrist or ankle joint; 3, swelling in the entire paw; and 4, deformity and or ankylosis. The maximum score was 16 per mouse. Two evaluators (one knew the group allocation, the other did not) independently scored arthritis and arrived at agreements on final scorings. At the end of the study, all mice were euthanized after giving anesthesia using 2% isoflurane (Patterson Veterinary, catalog number: 07–893-2374).
