Liposome Formulation with Tetraether Lipids

Curcumin (≥80%), cholesterol (≥99%), HEPES (≥99.5%), ethanol (absolute, ≥99.8%), D-(+)-trehalose dehydrate (≥99%) and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) were obtained from Sigma-Aldrich Chemie GmbH (Taufkirchen, Germany). The lipids 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) and 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE) were a gift from Lipoid GmbH (Ludwigshafen am Rhein, Germany). Fractions containing the polar tetraether lipids caldarchaeol (GDGT, approx. 10% of the extract) and calditoglycerocaldarchaeol (GDNT, approximately 90% of the extract) were extracted from the biomass of Sulfolobus acidocaldarius (Transmit GmbH, Gießen, Germany) [31 (link)]. The ends of GDNT contain phosphatidylmyoinositol and β-glucose respectively while GDGT contains phosphatidylmyoinositol and β-D-galactosyl-D-glucose on either ends respectively [32 (link)]. Ultrapure water from PURELAB® flex 4 (ELGA LabWater, High Wycombe, UK) was used for all experiments in this study. All solvents used were of analytical or HPLC grade.

Chitosan-Based Intranasal Desmopressin Formulation

Chitoceuticals chitosan 95/100 (degree of deacetylation 96%, Mw 100–250 kDa, chitosan-96) was purchased from Heppe Medical Chitosan (Halle, Germany). Polyethylene oxide (Mw 900 kDa, PEO), bovine serum albumin (BSA), acetic acid anhydride, Hank’s balanced salt solution (HBSS), Dulbecco’s phosphate buffered saline (PBS), glycerol (≥99%), tributyl citrate and ethyl cellulose, trifluoroacetic acid (TFA), and acetonitrile were obtained from Sigma Aldrich (St. Louis, MO, USA). N-2-hydroxyethylpiperazine-N’-2-ethanesulfonic acid (HEPES) was obtained from PanReac AppliChem (Darmstadt, Germany). Iron(III) oxide (Secovit® E172) was from BASF (Copenhagen, Denmark). Desmopressin as TFA salt (purity > 98%) was obtained from SynPeptide (Shanghai, China). MiniRin® freeze-dryed tablets-60 µg desmopressin (Ferring Pharmaceuticals, Copenhagen, Denmark) were purchased through the Association of Danish Pharmacies. Ultrapure water (18.2 MΩ × cm) purified by a PURELAB flex 4 (ELGA LabWater, High Wycombe, UK) was used.

Curcumin-Loaded PLA Nanoparticles for Biomedical Applications

PLA (molecular weight (Mw) = 420,000 g/mol, polydispersity = 1.5) was purchased from NatureWorks LLC (Minnetonka, MN, USA). ICG was ordered from Carl Roth GmbH (Karlsruhe, Germany). Curcumin (95%) was purchased from Alfa Aesar (Karlsruhe, Germany). Chloroform was purchased from Acros Organics (Geel, Belgium), and methanol and Tween80 were ordered from VWR (VWR International, Radnor, PA, USA). 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT), dimethylsulfoxide (DMSO), Triton X, and Uric acid were purchased from Sigma Aldrich Chemie GmbH (Taufkirchen, Germany). Ultrapure water was used for all the experiments. It was generated by PURELAB® flex 4 equipped with a point-of-use biofilter (ELGA LabWater, High Wycombe, UK).

Curcumin-loaded PLGA-DPPC-PEG Nanoparticles

Curcumin (95%) was purchased from Alfa Aesar (Kandel, Germany). PLGA (Purasorb PDLG 5002A) was obtained from Corbion Purac (Amsterdam, the Netherlands). The 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) and 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy(polyethylene glycol)-2000] (DSPE-PEG2000) were ordered from Avanti Polar Lipids (Alabaster, AL, USA). The 2′,7′-dichlorofluorescein diacetate, tetramethylrhodamine ethyl ester (TMRE), and carbonyl cyanide 4-(trifluoromethoxy)phenylhydrazone (FCCP) were purchased from Abcam (Cambridge, UK). The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), poly(vinyl alcohol) (PVA, Mowiol 4–88), tert-butyl hydroperoxide (TBHP), 2′,7′-dichlorofluorescein diacetate (DCFH-DA), dimethyl sulfoxide (DMSO), and acetonitrile were all supplied by Sigma-Aldrich Chemie GmbH (Taufkirchen, Germany). Acetone was acquired from Fisher Scientific GmbH (Dreieich, Germany). Ultrapure water (PURELAB flex 4, ELGA LabWater, High Wycombe, UK) was used in all experiments.

Synthesis of Bolalipid Compounds

The bolalipids dotriacontane-1,1′-diylbis [2-(trimethylammino)ethyl phosphate] (PC-C32-PC) and dotriacontane-1,1′-diylbis [2-(dimethylammino)ethyl phosphate] (Me2PE-C32-Me2PE) were synthesized at the MLU Halle-Wittenberg (Halle, Germany) according to procedures described previously [40 (link),41 (link)]. Sodium dodecyl sulfate and methylene blue were purchased from Carl Roth GmbH and Co. KG (Karlsruhe, Germany). Hydroxyethyl cellulose 300 (HEC 300; Mw ~ 807 g/mol) was obtained from Caesar and Loretz GmbH (Hilden, Germany). Physiological saline solution was purchased from B Braun (Melsungen, Germany). Saccharomyces cerevisiae were obtained from a retail outlet. Clinically isolated Staphylococcus aureus (ATCC 25923) was determined using MALDI Biotyper (Bruker Corporation, Billerica, MA, USA). Fertilized chicken eggs were obtained from Brormann GmbH (Rheda-Wiedenbrück, Germany). For all experiments ultrapure water from PURELAB®® flex 4 (ELGA LabWater, High Wycombe, UK) was used.

Potentiometric Sensor Membrane Fabrication

Tetrahydrofuran (THF) and high molecular weight poly(vinyl chloride) (PVC), potassium tetrakis(4-chlorophenyl)borate (KTpCIPB), ammonium ionophore I (nonactin), bis(2ethylhexyl) sebacate (BEHS), and dibutyl phthalate (DBP) tetradodecylammonium nitrate (TDDAN) were purchased from Sigma-Aldrich and used without further purification in the preparation of the membranes. Graphite (Sigma-Aldrich, USA), epoxy (Macroplast Su 2227, Henkel, Germany), and hardener (Desmodur RFE, Bayer AG, Germany) were used in the preparation of the solid contact material. All cationic and anionic standard solutions were prepared using analytical grade nitrate or chloride salts, sodium, or potassium salts, respectively. Ultrapure water (18.2 MΩ cm -1 ) was obtained via an ultrapure system Purelab Flex 4 (ELGA LabWater, UK). All ISE potentials were measured against the silver/silver chloride reference electrode, which was elaborated in our laboratory.

Solubilization of Parietin using Cyclodextrin
