For miRNA qRT-PCR (Quantitative reverse transcription PCR) analyses, miRNA first-Strand was synthesized using the Mir-X™ miRNA First-Strand Synthesis and SYBR® qRT-PCR Kit according to the instructions (638,313, Takara Biotechnology Co., Ltd). For target mRNA qRT-PCR analyses, first-strand cDNA was synthesized using TransScript® One-Step gDNA Removal and cDNA Synthesis SuperMix (AT311, Transgene, Beijing, China) (Pei et al. 2020 (link)).
The qPCR experiment was performed on LightCycler 480 Real-Time PCR instrument (Roche Diagnostics®, Basel, Switzerland). All reactions were performed with three replications, and U6 snRNA and PyTUB were respectively used as the internal control for miRNA and mRNA. Relative expression level of each gene was calculated using the 2-ΔΔCp algorithm. The qPCR primers are listed in Supplementary Table
