Deionized water was purified with a Millipore Milli-Q RG water purification system (Darmstadt, Germany). Dithiothreitol and iodoacetamide were purchased from Thermo Fisher Scientific (Waltham, MA) and freshly prepared in 50 mM ammonium bicarbonate buffer each day before use. MS-grade trypsin and Lys-C were products of Promega (Madison, WI). Unless otherwise mentioned, all other reagents were obtained from Sigma-Aldrich (St. Louis, MO).

Preparation and Characterization of IMQ-Loaded Lipid Formulations

Imiquimod (IMQ, purity > 98%, molar mass 240.31 g/mol) and oleic acid (OA, >99%) were purchased from TCI chemicals (Tokyo, Japan). Phospholipids (PL) from egg yolk (60% phosphatidylcholine, mostly 16:0/18:2, 15% phosphatidylethanolamine, mostly 18:0/18:2 and 18:0/20:4, minor fractions phosphatidylinositol, lysophosphatidylcholine, sphingomyelin, lysophosphatidylethanolamine), acetic acid (>99%), ethanol (>99%), acetonitrile (HPLC grade), and methanol (HPLC grade) were purchased from Sigma Aldrich (Schnelldorf, Germany). Aldara topical cream (5% IMQ) MEDA company, Sweden was obtained from Hospital Pharmacy, Hradec Králové, Czech Republic. Water was deionized, distilled, and filtered through a Milli-Q RG water purification system (Millipore, Burlington, MA, USA).

Cyclodextrin Capillary Electrophoresis Analysis

All chemical and solvents used were of analytical grade or of HPLC quality.
β-CD was supplied by Sigma (Saint-Louis, MO, USA). α-CD, γ-CD, and dimethyl-β-CD (Me-β-CD) were purchased from Acros Organics (Fisher Scientific, IIIkirch, France) and 2-hydroxypropyl-β-CD (HP-β-CD) was from Cyclo Lab LTD (Budapest, Hungary). The tetraborate sodium buffer (10 mM, pH = 9.5) was prepared from Na2B4O7.10H2O (VWR, Fontenay-sous-Bois, France). Different concentrations of CDs were prepared with this tetraborate buffer for ACE experiments. The phosphate buffer (40 mM, pH = 7.0) was prepared by mixing appropriate amounts of NaH2PO4 (Carlo Erba, Val-de-Reuil, France) and Na2HPO4
(Sigma, Saint-Louis, MO, USA).
A thiourea solution at 0.01% (w/w) in water was used as an electroosmotic flow (EOF) marker. thiourea was from VWR (Fontenay-sous-Bois, France).
A MilliQ RG water purification system (Millipore, Bedford, MA, USA) was used to provide ultrapure water (18.2 MΩ.cm).
All buffers were filtered through a 0.20 µm nylon membrane from VWR supplier.
