Hard gelatin capsules (size 1) were purchased from Capsugel Inc., Morristown, NJ, USA, and all other reagents used are of analytical grade.

Comprehensive Lipid Extraction and Quantitation

Approximately 4–5 × 106 MPMs were subjected to monophasic lipid extraction in methanol:chloroform:water (2:1:0.74, v:v:v) as previously described [46 (link),79 (link)]. Pelleted cellular proteins were subsequently dissolved in 0.1 N NaOH to determine the total amount of protein per sample group and used for normalization of lipids detected. Di-myristoyl phosphatidylcholine (Avanti Polar Lipids, Alabaster, AL, USA) and 19-hydroxycholesterol (Steraloids, Newport, RI, USA) were added to samples during lipid extraction at 1 nmol/mg protein as an internal standard for relative quantitation of lipids and sterols. Dried lipid extracts were desalted by washing as previously described and resuspended using 200 µL/mg protein in a solution of methanol containing 0.01% butylated hydroxytoluene [79 (link)]. The samples were stored under a blanket of nitrogen at −80 °C until further analysis.

Overexpression of MSP1E3D1 in E. coli

Lipid Vesicle Encapsulation of Dual-Labeled DNA

We encapsulated two kinds of TA-rich DNAs, one labeled with Cy3 and the other labeled with Cy5/Cy7 at both ends, in lipid vesicles as described earlier (6 (link)). Briefly, lipid films were prepared by mixing 1,2-dipalmitoyl-sn-glycero-3-phosphoethanolamine-N-(Cap Biotinyl) (0.05 mg/ml) with dimyristoyl phosphatidylcholine (5 mg/ml) (Avanti Polar Lipids) in chloroform and gently drying with nitrogen gas in glass bottle. The lipid film was hydrated with 100 μl buffer solution containing 100 mM NaCl and 25 mM Tris–HCl pH 8.0, followed by repeated freeze-thaw cycles to homogenize the suspension. Two kinds of DNAs were added to make total volume of 200 μl and concentration of 400 nM for each, which corresponds to one molecule in a spherical volume of 200 nm diameter. The solution was extruded 11 times through membrane filter with 200 nm track-etched pores (Whatman, 800281) to create uniformly sized vesicles. The encapsulation efficiency of 15 ± 3% was estimated as the ratio of the number of vesicles containing two dyes (two Cy3’s, two Cy5’s, or one of each Cy3 and Cy5) to the number of vesicles containing one dye (Cy3 or Cy5).

Recombinant FVIII Characterization

Preparing Lipid Samples with Cosolvents

The phospholipids dimyristoylphosphatidylcholine (DMPC) and palmitoyl-oleoyl-phosphatidylcholine (POPC) were purchased from Avanti Polar Lipids, Inc. (Alabaster, AL). Urea and TMAO were obtained from Sigma-Aldrich and NaCl from VWR. The samples were prepared so that the co-solute-lipid mass fraction (of urea, TMAO or NaCl) in the dry samples, C lip cosol = m cosol /(m cosol + m lip ), was fixed while the water content in terms of the number of water molecules per lipid n w = N w /N lip is variable. The samples were mixed together by the use of a mortar and pestle to produce a fine powder, which was then subjected to drying under vacuum with a 3 Å molecular sieve for at least 24 hours. According to earlier reports, 27, 28 this drying procedure removes all water from the lipid sample. The dry samples were then loaded into the measurement device under a stream of dry N 2 .
