TBXA2R is a laboratory measurement device for the quantitative detection and analysis of thromboxane A2 receptor (TBXA2R) protein expression levels in biological samples. The core function of TBXA2R is to provide accurate and reliable data regarding the presence and quantity of TBXA2R, which is a key regulator of various physiological processes.

Quantification and Visualization of Protein Levels

Check if the same lab product or an alternative is used in the 5 most similar protocols
The BCA assay quantified proteins, and loading buffer 5X was added to the proteins, which were incubated for 5 min at 95°C. Then, proteins were loaded on an SDS-PAGE polyacrylamide gel, transferred to Immobilon-P PVDF membrane (Millipore, Shanghai, China), probed with the appropriate primary antibodies: COX-1 (1:5000), COX-2 (1:4000), and PTGFR (1:2000) were obtained from Abcam (Cambridge, UK), and TBXA2R (1:4000), GAPDH (1:10000), and detected by chemiluminescence (ECL, Thermo Scientific). Images were then acquired with Image-Lab software (Bio-Rad, Hercules, CA, USA). Image analysis of western blots was performed with Image-Lab analyzer software.

Protein Expression Analysis of Vascular Segments

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cleaned from connective and fat tissue, supraceliac, paravisceral, and juxtarenal segments were stored at −80°C. For WB analysis, 5–6 segments per group were pulled, homogenized in RIPA buffer, and lysates were analyzed with sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and immunoblotting. The following primary antibodies were used for WB analysis: α1A-AR (#sc-100291, SCBT, TA, USA), TBXA2R (#PA5-67751, Thermo Scientific, Waltham, MA, USA), and β-actin (#A2228, Sigma, St. Louis, MO, USA).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
