Cobalt protoporphyrin copp

Manufactured by Enzo Life Sciences
Sourced in United States

Cobalt protoporphyrin (CoPP) is a metalloporphyrin compound used as a research tool in cell biology and biochemistry laboratories. It is a cobalt-containing analog of the natural compound protoporphyrin IX. CoPP can be utilized in various experimental setups to study cellular processes, signaling pathways, and the effects of metal-containing porphyrins on biological systems.

Check if the same lab product or an alternative is used in the 5 most similar protocols
Crocin (Figure 1(a)), LPS (phenol extracted from Salmonella enteritidis), 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), actinomycin D (Act. D), cycloheximide (CHX), 2-(3,6-bis(acetyloxy)-2,7-dichloro-9H-xanthen-9-yl)benzoic acid acetoxymethyl ester (Fluo-3/AM), and other reagents were purchased from Sigma-Aldrich (St. Louis, MO, USA). Cobalt protoporphyrin (CoPP) and zinc protoporphyrin IX (ZnPP) were bought from Enzo Life Sciences, Inc. (Farmingdale, NY, USA). LY294002, PD98059, SP600125, and SB203580 were obtained from A.G. Scientific (San Diego, CA, USA). Nifedipine, 1,2-bis (o-aminophenoxy) ethane-N,N,N′,N′-tetraacetic acid tetra (acetoxymethyl) ester (BAPTA/AM), and calmidazolium chloride were acquired from Calbiochem (EMD Millipore, Billerica, MA, USA). KN92, KN93, and KN62 were purchased from Cayman Chemical (Ann Arbor, MI, USA) and Calbiochem.

HEK293 Cell Culture and PAO Treatment

Check if the same lab product or an alternative is used in the 5 most similar protocols
Human embryonic kidney (HEK293) cells were purchased from the American Type Culture Collection (CRL-1573) and maintained in MEM medium (HyClone, Logan, UT) containing 10% fetal bovine serum (Sigma, St. Louis, MO), 1% antibiotic/antimycotic solution (Mediatech, Manassas, VA) at 37 °C in a 5% CO2 incubator. PAO (Sigma) stock solution was freshly prepared for each exposure in 100% DMSO as described previously.14 (link) PAO treatment of HEK293 cells was done employing various concentrations (0–250 nM) diluted in the culture medium. Vehicle (alone)-treated cells served as controls. Cobalt protoporphyrin (CoPP) (Enzo Life Sciences, Farmingdale, NY), an inducer of HO-1, was used in some experiments as described in the figure legends. In these experiments, cells were pretreated with CoPP (10 μM) for 6 h before PAO challenge and then both treatments were continued for 24 hours.

Anti-inflammatory Effects of Dioscorea moniliforme Extract

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cobalt protoporphyrin (CoPP) was purchased from Enzo Life Sciences, Inc. (USA). Actinomycin D (AD), cycloheximide (CHX), 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), and LTA from Staphylococcus aureus were purchased from Sigma-Aldrich Co. (USA). LY294002, PD98059, SP600125 and SB203580 were obtained from A.G. Scientific (USA). The antibodies for iNOS, HO-1, Nrf2, Akt, p38, TATA-box binding protein (TBP) and histone deacetylases (HDAC) were purchased from Santa Cruz Biotechnology (USA), the antibodies for phospho-Akt and phospho-p-38 were purchased from Cell Signaling Technology (USA), and the anti-tubulin antibody was purchased from Bio Genex (USA). The dry shoot systems of D. moniliforme Sw. were obtained and extracted as described previously [30] . In brief, the dry shoot systems (300 g) were extracted with distilled water at 100ºC for 4 h. The extract was filtered through a 0.45-μm filter, freeze-dried (yield: 8 g), and stored at 4ºC. The dried extract was dissolved in phosphate-buffered saline (PBS) and filtered through a 0.22 μm-filter before use.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
