Retroviral pmx vector

Manufactured by Cell Biolabs

The Retroviral pMX vector is a plasmid-based expression system designed for the production of recombinant retroviruses. The vector contains the necessary genetic elements for viral packaging and gene delivery, allowing for the efficient transduction of target cells with the gene of interest.

Transient Expression of mDia2 and ATP Biosensors in Human Fibroblasts

Check if the same lab product or an alternative is used in the 5 most similar protocols
Wild-type or mutant Flag-tagged mDia2 cDNAs (17, 20 (link)) were subcloned into the retroviral pMX vector (Cell Biolabs). pGEM-MIRO1 and cyto-Ruby3-iATPSnFR1.0 (ATP biosensors; ref. 21 (link)) plasmids were purchased from Sino Biological Inc. (#HG15898-G) or Addgene (#102551), respectively. Plasmid vectors and Lipofectamine 2000 (#11668030, Invitrogen) were incubated with Opti-MEM I Reduced Serum Medium (#31985062, Thermo Fisher Scientific) at room temperature for 20 minutes. The mixture was then added to primary human fibroblasts. After 6 hours of incubation, the medium was replaced by a normal culture medium containing 10% FBS. Cells were allowed to recover for 24–48 hours prior to analysis.

Overexpression of Flag-mDia2 in Fibroblasts

Check if the same lab product or an alternative is used in the 5 most similar protocols
Wild‐type Flag‐tagged mDia2 cDNA (Isogai et al, 2015a, 2016) was subcloned into the retroviral pMX vector (Cell Biolabs). One microgram pMX Flag‐mDia2 or empty vector and Lipofectamine™ 2000 (#11668030, Invitrogen) were incubated with Opti‐MEM™ I Reduced Serum Medium (#31985062, Thermo Fisher Scientific) at room temperature for 20 min. The mixture was then added to primary human fibroblasts. After 6‐h incubation, medium was replaced by normal culture medium containing 10% FBS. Cells were allowed to recover for 48 h prior to analysis.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
