Quantifying Viral Titers in Swabs

Challenge dose and infectious virus in swab samples were determined by agarose plaque assay, run on fresh (i.e. not frozen then thawed) samples the day of collection. Required dilutions of each specimen were prepared in virus diluent [1X minimum essential media (Corning Life Sciences, Pittston, PA) containing 10% fetal bovine serum (Cytiva Life Sciences, Marlborough, MA), 1% GlutaMAX (Gibco, Waltham, MA), and 1% NEAA (Sigma, St. Louis, MO)] in duplicate (swab specimens) or triplicate (all-glass impinger specimens from Study Day 1), were added to plates containing Vero 76 cells (ATCC, Manassas, VA) at greater than or equal to 85% confluency. Two days later, the cells were stained with neutral red (Thermo Fisher Scientific, Waltham, MA), and plaque counts were obtained the day after staining.

Culturing Jeg-3 Cells in MEM

Jeg-3 cells were purchased from the American Type Culture Collection (Manassas, VA) and maintained in minimum essential media (Corning Life Sciences) supplemented with 10% FBS (Genesee Scientific) and 100 μg/mL Streptomycin, 100 U/mL Penicillin (HyClone), and humidified in 5% CO2 at 37°C.

Cell Culture and GABAergic Differentiation

Cell culture was performed as described by us previously 6 (link), 7 (link), 9 (link), 39 (link), 40 (link). Briefly, HeLa, HEK293T and HepG2 cells were maintained in minimum essential media (Corning, Corning NY) supplemented with 10% fetal bovine serum (Atlanta Biologicals, Flowery Branch, GA), 100 IU/ml penicillin-G, and 100 μg/ml streptomycin (both from Corning). AF5 cells were grown in Dulbecco’s modified Eagle’s medium/Ham’s F-12 (Life Technologies) with 10% fetal bovine serum, 2 mm L-glutamine, 100 IU/ml penicillin-G, and 100 μg/ml streptomycin (maintenance media). GABAergic differentiation of AF5 cells was performed by transferring cultures to Neurobasal media supplemented with serum-free human B27 (Life Technologies) and 2mM L-glutamine for 48–72 h, as described by us previously 6 (link).
