Ox-LDL-Induced Atherosclerosis Model in EA.hy926 Cells

Human umbilical vein EA.hy926 cells (Cell bank, Shanghai Institute for Biological Science, Shanghai, China) were cultured in Dulbecco's modified Eagle medium (DMEM; Invitrogen, CA, USA) supplemented with 10% heat-inactivated fetal bovine serum (FBS; Invitrogen, CA, USA) at 37°C in a humidified atmosphere of 95% air and 5% CO2. Cells were passaged according to the number of cells required for different experiments. The second day after passage, the cells were incubated with 10, 100, 250 and 500 µg/ml ox-LDL (Yuanye Biotechnology, Shanghai, China) and/or with 500 µM salidroside (purity, >98%; Yuanye Biotechnology; 50 µM pravastatin was used as a positive control) for 48 h to establish lipid oxidation and AS therapy models. Then, the cells were incubated with 20 µl of CCK-8 (Beyotime, Shanghai, China) for 2 h, according to the manufacturer's instructions. Cell proliferation was analyzed using a microplatereader, and the absorbance was measured at 450 nm (Miltenyi Biotec GmbH, Bergisch Gladbach, Germany).

Comprehensive HPLC Analysis of Medicinal Metabolites

Reference compounds of HPLC grade, i.e., salidroside, calycosin-7-O-β-D-glucoside, specnuezhenide, formononetin, and polyphyllin I, were purchased from Shanghai Yuanye Bio-Technology Co., Ltd. with purity ≥98%. Puerarin as an internal standard substance (IS) was obtained from the National Institutes for Food and Drug Control (Beijing, China). Figure 1 lists the chemical structures of these metabolites. Distilled and deionized water was prepared with a water purification system (RODI-220A1, RSJ, China).
HPLC-grade acetonitrile and methanol were obtained from AQA (Cleveland, United States) and Guangzhou Chemical Reagent Factory (Guangzhou, China). The other chemicals and reagents were of analytical grade and were commercially available.

Herbal Compound Treatment for Alzheimer's

Amyloid beta A4 rabbit antibody (Catalog No: A17911) and GSK3β rabbit antibody (Catalog No: A11731) were purchased from Wuhan ABclonal Biotech Co., Ltd. The standard products of salidroside, umbelliferone, gastrodin, and nicotinamide were purchased from Yuanye Biotechnology Co., Ltd. (China). Acetonitrile and methanol were supplied by Merck KGaA (Darmstadt, Germany). Formic acid of chromatographic grade was purchased from Aladdin (Shanghai, China). Ultrapure water was prepared by a water purification system (Milli-Q Integral 5). All other chemicals were of analytical grade.

Salidroside Effect on Cigarette Smoke-Induced COPD

The rats were randomized and treated intraperitoneally (IP) with vehicle (sterile injectable water) or Salidroside (Shanghai Yuanye Bio-Technology, Shanghai, China) at 50 mg/kg (COPD+50 mg/kg group), 100 mg/kg (COPD+100 mg/kg group) or 200 mg/kg (COPD+200 mg/kg group), followed by cigarette smoking (Hongmei cigarettes, 13 mg CO, 13 mg Tar oil) for 30 min/day, twice per day at an interval of 6–8 h, 6 days per week for 16 weeks in an 8050™ inhalation exposure system (6–8% of smog concentration, Tianjin, China) [24–27 (link)]. A healthy control group of rats received vehicle injection and were exposed to normal air (n=8 per group).

Salidroside Inhibits Tumor Growth in Nude Mice

Eighteen nude mice (Shanghai Laboratory Animal Company) were subcutaneously injected with BxPc‐3 cells (4 × 106 cells, 100 μL) and randomly divided into three groups: control group, salidroside (25 mg/kg) group, and salidroside (50 mg/kg) group. After tumors formation (approximately 1‐2 weeks later), tumor volume (mm3) was calculated every 3 days from 12th day to 39th day. When the tumor volume reaching 100 mm³ , mice in the control group was normal fed without any treatment, mice in two salidroside groups were intragastrically administrated with salidroside (B20504,YuanYe Biotechnology (Shanghai) Co., Ltd, Shanghai, China) at a dose of 25 mg/kg and 50 mg/kg, respectively. Three weeks later, all the mice were killed. Tumor weight were calculated and metastasis in lung was assessed using hemotoxylin and eosin (H&E) staining according to a reported study.20 We confirmed that the use of animal was approved by animal ethics committee of Shuguang Hospital affiliated to Shanghai University of Traditional Chinese Medicine.

Salidroside characterization and MTT assay

Salidroside (CAS registry number: 10338-51-9, batch number: Y18J8Y17024) was purchased from Yuan Ye Biotech. (Shanghai, China). According to the manufacturer's data, the purity was 98% based on HPLC analysis. The chemical structure is shown in Figure 1(a). 3-[4,5-Dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) was purchased from Sigma-Aldrich (St. Louis, MO, USA).

Fingerprinting SHTL Powder Compounds

Based on previous reports, 10 different batches of SHTL powders were separated using a ZORBAX SB-C18 column (4.6 × 250 mm, 5 μm, Agilent, Santa Clara, CA, USA) and analyzed for chemical fingerprints using high-performance liquid chromatography (HPLC, Shimadzu Corp., Nakagyo-ku, Kyoto, Japan) coupled with a diode array detector (DAD) (Shimadzu Corp.) [21 (link)]. Acetonitrile in water (solvent A) and 0.4% phosphoric acid in water (solvent B) constituted the mobile phase (Supplementary Table 1). The flow rate was 1.0 mL/min at 25°C, and the detection wavelength was set at 203 nm. Salidroside, chlorogenic acid, paeoniflorin, ferulic acid, luteoloside, ginsenoside Rg1, and luteolin were used as controls to analyze the retention time (Shanghai Yuanye Biotechnology Co., Ltd.; Shanghai, China).
