Molecular Characterization of Drug Resistance in Falciparum Malaria

Skin Snip DNA Extraction and Filarial PCR

Preparation of the DNA template: skin snip biopsies DNA were extracted using the QIAamp DNA Mini Kit with a incubation at 56°C overnight previous to DNA extraction. DNA, was eluted with 200 μl distilled water and stored at 4°C until use.
Detection of filarial species was performed by Real Time PCR, from a modification of a Nested Filaria PCR [20 (link)]. The amplified products were purified for sequencing using Illustra DNA and Gel Band Purification Kit (General Electric Healthcare, England) and sequenced using Big Dye Terminator v3.1 Cycle Sequencing in an ABI PRISM 3700 DNA Analyzer (Applied Biosystems, U.S.A.).
