Islet ATP and IP3 Measurement

After preincubation at 2.8 mM glucose for 60 min, groups of ten islets were incubated in tubes containing 0.5 ml KRBH buffer supplemented with 2.8 or 11 mM glucose with or without 300 nM CNX-011-67 for 60 min for ATP and 5 min for IP3 measurement. The islets were then washed twice with 1X cold PBS and lysed in lysing buffer provided by the manufacturer.
