Optimized Protocol for In Vivo and Ex Vivo Drug Delivery

Um-PEA (0.8–6.0 μm) was kindly provided by EPITECH Group SpA, Saccolongo (PD). MPEP hydrochloride was purchased from Tocris Bioscience (Bristol, UK). MDCPG was purchased from Hello Bio (Bristol, UK). Drugs were dissolved in 0.05% dimethylsulfoxide in artificial cerebrospinal fluid (ACSF) on the day of the experiment. Pharmacological Modulation of Microglial Lipid Signaling

To block voltage-gated sodium channels, 1 μM Tetrodotoxin (Cat#1078; Tocris) was applied to the media before electrostimulation of spheroids. Control cells were treated with DMSO as vehicle. To induce lipid accumulation, 20 μM Oleic Acid (Cat# 03008; Sigma-Aldrich) was applied overnight to iMGLs, control cells were treated with 0.1% BSA as vehicle. 1 μM Triacsin C (Cat#10007448; Cayman Chemical) was applied to iMGLs overnight to deplete lipid accumulation.

Pharmacological Modulation of Synaptic Plasticity

N-[2-(4-Chlorophenyl)ethyl]-1,3,4,5-tetrahydro-7,8-dihydroxy-2H-2-benzazepine-2-carbothioamide (capsazepine, 10 μM; Tocris) and (E)-N-[(4-hydroxy-3-methoxyphenyl)methyl]-8-methyl-6-nonenamide (capsaicin, 10 μM; Tocris) were dissolved in DMSO and then added in the external solution at a final concentration of DMSO of 0.001 and 0.0025, respectively. Tetrahydrolipstatin (THL, 10 μM; Sigma) was dissolved in DMSO (0.08%) and applied internally via the patch-clamp pipette. DMSO (0.08%), the vehicle used to dilute i-THL, did not preclude the tLTP induced with 10 post-pre pairings (Cui et al., 2015 (link)).
