Quantitative RT-PCR Analysis of STAT5 Target Genes

RNA was extrated from untreated and IMB-HDC-treated cells expressing wild-type or mutant STAT5 variants using Trizol reagent (Invitrogen, Waltham, MA, USA). cDNA synthesis was performed with Oligo (dT) or random primers by using a Quantscript RT Kit (Tiangen). Real-time PCR was performed with SsoAdvanced SYBR GreenSupermix (Bio-Rad) and primers for Bcl-2 [10 (link)] and c-myc (forward 5′-AACAGGAACTATGACCTCG-3′, and reverse 5′-AGCAGCTCGAATTTCTTC-3′), CyclinD1 (forward 5′-ACAAACAGATCATCCGCAAACAC-3′ and reverse 5′- TGTTGGGGCTCCTCAGGTTC-3′), Pim-1 (forward 5′-TCTTCTGGCAGGTGCTG-3′ and reverse 5′-GGTAGCGAATCCACTCTG-3′), Osm (forward 5′-AGATACCTGAGCCCACACAGACAG-3′ and reverse 5′-ATCGTCCCATTCCCTGAAGACC-3′), and c-Fos (forward 5′-CGAAGGGAACGGAATAAGATGG-3′ and reverse 5′-AGACCTCCAGTCAAATCCAGGG-3′). Primers for target gene expression of STAT5 were normalized to GAPDH.
