Nano-IL-12 Therapy in BALB/c Mice

Healthy female BALB/c mice (5 weeks old) were i.v. injected twice with 10 µg IL‐12 or equivalent Nano‐IL‐12 on Day 0 and Day 3 with daily recording the bodyweight. On Day 7, mice were sacrificed for harvesting blood, liver, and kidney samples. The blood samples were kept under room temperature for complete coagulation, followed by centrifugation at 4000 g for 10 min to collect serum. The serum samples were loaded to blood chemistry analyzer (DRI‐CHEM NX700, Fujifilm) to detect the concentration of total protein (TP), alanine aminotransferase (ALT), blood urea nitrogen (BUN), lipase and creatinine (CRE). The liver and kidney samples were embedded in O.C.T compound and frozen by liquid N2. The tissues were sliced by cryostat (CM1950, Leica) to 10 µm thickness sections. The sections were stained by H&E kit and observed by microscope (BZ‐X810, Keyence) with brightness field.

Blood Collection and Biochemical Analysis

Before imaging, blood samples were collected from all rats under anesthesia. Using a heparinized capillary tube (Marienfeld HSU‐29 00 000, Germany), 0.5–1 mL of blood was drained from the retro‐orbital sinus and stored at room temperature for 30 min to induce coagulation. For diabetic experiments, the BG level was measured on the spot with a glucometer (Accu‐Chek Guide Set, Roche Diabetes Care GmbH, Germany). The sampled blood was microcentrifuged for 10 min at 10 000 g and 4 °C, and the supernatant (serum) was separated and stored in a deep freezer at below −80 °C. The sCr and BUN concentrations were measured from thawed blood serum samples using an automatic biochemical analyzer (DRI‐CHEM NX700, Fujifilm, Japan) with paired biochemical test slides (DRI‐CHEM SLIDE, Fujifilm, Japan).

Serum Toxicity Profiling of Combination Therapies

To evaluate the serum toxicity profile of combination therapy regimens, an A549 human xenograft tumor model was established by subcutaneously injecting 1 × 107 cells into 6- to 8-week-old male athymic nude mice (Orientbio). Assessing Renal Function in Mice
