Cuprizone

Manufactured by Thermo Fisher Scientific
Sourced in United States

Cuprizone is a laboratory chemical compound used in scientific research. It is a copper chelator that has been utilized in various studies, particularly in the field of neuroscience. The core function of Cuprizone is to induce demyelination, a process in which the protective myelin sheath surrounding nerve fibers is damaged or removed. This research tool is commonly employed to create animal models for the study of demyelinating diseases, such as multiple sclerosis.

Cuprizone-Induced Demyelination and Ponesimod Treatment

Check if the same lab product or an alternative is used in the 5 most similar protocols
All animal procedures were conducted in accordance with Institutional Animal Care and Use Committee guidelines of Sanford Burnham Prebys Medical Discovery Institute. C57BL/6J 8-week-old male mice were maintained on a diet of 0.2% cuprizone (Bis(cyclohexanone) oxaldihydrazone 98%; Thermo Fisher Scientific, Cat # 370-81-0) for 5-6 weeks. A suspension of ponesimod (30 mg/kg) in vehicle (0.25% methylcellulose (400 cp) with 0.05% polysorbate 80) was prepared weekly after stirring overnight at room temperature and aliquoted accordingly to store at 4°C. Vehicle or ponesimod were delivered orally twice a day (8 hr apart) by re-equilibrating the suspension upon stirring for 30 minutes before the procedure. The ponesimod dose, frequency, delivery, and formulation was based on a prior study published at ACTRIMS.

Fluorescein-labeled Peptide Synthesis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Peptides were synthesized with an N-terminal fluorescein amine (FAM) tag and provided at >90% purity by (Genscript). Lyophilized aliquots of ~3–4 mg were stored at −80°C and reconstituted with sterile phosphate-buffered saline (PBS) for use in mice. Lysolecithin was from Sigma (Cat# L4129) and cuprizone from Thermo Fisher Scientific (Cat# 370-81-0).

Cuprizone and Palmaria Effects on Body Weight

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cuprizone (Thermo Fisher Scientific-C9012-25G; Waltham, MA, USA) was freshly prepared daily and blended with ground regular chow in 0.2% w/w of CPZ [66 (link)]. Palmaria p. leaf Powder Liquid Extract 2 × 4 oz was purchased from HawaiPharm LLC (Honolulu, HI, USA). The effect of CPZ and Palmaria p. on body weight was measured by the percentage of body weight gain throughout the seven weeks of the experiment following this equation: weight gain (%) = (new weight [W1] − initial weight [W0]/initial weight [W0]) × 100 [67 (link)].

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
