Acetaminophen Metabolites and Methemoglobinemia

The effect of acetaminophen metabolites on methemoglobinemia was tested in vitro by adding them to a sample of perfusate (1:1 whole blood with modified Krebs-Henseleit). P-aminophenol (Sigma Aldrich Canada, Oakville, ON, Canada) was added in a concentration of 100 µg/mL and let sit at room temperature. Methemoglobin was measured, as described above, at 30 and 90 min. N-acetyl-p-benzoquinone imine (NAPQI) (Sigma Aldrich Canada, Oakville, ON, Canada) was added to a separate sample of perfusate at a concentration of 125 µg/mL and methemoglobin was again measured after 90 and 180 min at room temperature. For comparison, acetaminophen (Sigma Aldrich Canada, Oakville, ON, Canada) was added to perfusate at a concentration of 10 mg/mL and methemoglobin was measured after 60 min. The effect of CCl4 on hemolysis was tested in vitro by adding CCl4 (Sigma Aldrich Canada, Oakville, ON, Canada) to whole blood in a concentration of 10 µL/mL and measuring plasma free hemoglobin after 30 min at room temperature, as detailed above. Free hemoglobin was measured in porcine whole blood after 30 min at room temperature without the addition of CCl4 for control comparison.

Synthesis and Characterization of Benzoxazine

Analytical Reagents for Pharmaceuticals

Daunorubicin Extraction Protocol

A stock solution of DNR at 1000 mg L−1 was prepared in 1 % (v/v) formic acid in water:methanol (50:50 % v/v). Working solutions were prepared by daily dilution of the stock solution.

85 w/w% phosphoric acid (ACS grade) was purchased from Fischer Chemical
(Hampton, New Hampshire). Potassium nitrate (reagent grade) was purchased
from VWR Life Sciences (Radnor, Pennsylvania). A 1,4-dimethyoxybenzene
derivative (C7) was synthesized40 (link) and provided
by the Zhang group from Argonne National Laboratory. A tri(dialkylamino)cyclopropenium
derivative (HS-cyclopropenium) was synthesized41 (link) and provided by the Sanford group from the University of
Michigan, Ann Arbor. Eighteen MΩ DI water was obtained from
a Milli-Q purification system. All other chemicals were used as purchased
without further purification. Glass slides (2” × 3”)
were purchased from Corning Inc. (Corning, New York). Anisotropically
conductive film for making device connections was purchased from 3
M (St. Paul, Minnesota). Photomasks for the device were purchased
from Front Range Photomasks (Phoenix, Arizona).

Adsorption of Paracetamol onto Modified Clay

All chemicals were of analytical grade. The clay used was Wyoming Na-montmorillonite SWY-2 obtained from the Source Clays Registry (Clay Mineral Society, Colombia, MO). Quartz sand (grain size 0.8-1.2 mm) was obtained from Negev industrial minerals (Israel). Octadecyltrimethylammonium (ODTMA) bromide was obtained from Sigma Aldrich. Paracetamol was obtained from Birzeit pharmaceutical company (Ramallah-Palestine). Activated Charcoal (12-20 mesh) was obtained from Sigma (Sigma Chemical Company, USA). De-ionized water was used to prepare all solutions. Methanol, acetonitrile, tetrahydrofuran, and water were both HPLC grade and purchased from Sigma Aldrich. Magnesium sulfate, hydrogen peroxide, and p-aminophenol were purchased from Sigma Aldrich. High purity diethyl ether (>99%) was purchased from Biolab (Israel); orthophosphoric acid was obtained from Riedel-De Haën (Germany). Microbiological growth media (MacConkey agar, Blood Agar, Eosin Methylene Blue (EMB) Agar, and Mannitol Salt Agar (MSA)) were obtained from Sigma-Aldrich company; API-20E test Kit was obtained from Biomerieux.
