Apoptosis Detection in Embryos

– after freezing embryos with liquid nitrogen, the embryos were fixed
with 4% PFA/PBS, pH 7.4 for 15 min at room temperature and then permeabilized by
incubation with 0.1% Triton X-100 in PBS for 10 min at room temperature. After
washing, TUNEL staining was performed according to the manufacturer’s
(Roche, Switzerland) instructions. Stained embryos were mounted using
Fluoroshield with DAPI mounting solution (Sigma-Aldrich) to visualize the
nuclei.
