Steady-State Kinetic Assay of MtuDAH7PS

Solutions of L-Phe (Sigma), L-Tyr (Sigma), and L-Trp (Sigma) in ultrapure water were added to standard MtuDAH7PS assay reaction mixtures. Inhibition study assays were conducted in the presence of none, one or two aromatic amino acids. MtuDAH7PS steady-state kinetic assay solutions contained PEP (150 μM), E4P (150 μM), MnSO4 (100 μM) and amino acid (0–200 μM) in assay buffer. For assays with two amino acids, the background concentration of the indicated amino acid was held at 100 μM. Reactions were initiated by the addition of MtuDAH7PS.

Analytical Preparation and Purification

All chemicals were of analytical grade and applied without further purification. Capillary Regenerator Basic Wash sodium hydroxide was purchased from Beckman Coulter (Brea, CA, USA). The water used in all experiments was obtained from Milli-Q equipment (Bedford, MA, USA).

Mushroom Tyrosinase Kinetics Characterization

Mushroom tyrosinase (EC 1.14.18.1) was the product of Sigma Chemical Co. (St. Louis, MO, USA) with specific activity of 6680 U/mg. Water used in this study was purified on a Millipore Milli-Q apparatus (TGI Pure 110 Water Systems, USA).

Authentic Chemical Compound Acquisition

Antioxidant Properties of Deer Velvet

Velvet antlers (Cervus elaphus Linnaeus) were obtained from male red deer that were bred at the Daxing’an Mountain range (Heilongjiang, China). Dulbecco’s modified Eagle’s medium and fetal bovine serum were purchased from Gibco BRL (Grand Island, NY, USA). Penicillin–streptomycin and NO Assay kit were purchased from Beyotime Biotechnology (Shanghai, China). HPLC-grade acetonitrile was purchased from Mallinckrodt Baker (Phillipsburg, NJ, USA). All other chemicals were of analytical grade unless otherwise mentioned.

Tyrosine Stereoisomer Preparation in PBS and aCSF

dissolved in Dulbecco’s PBS (122 mM NaCl, 3.0 mM KCl, 1.2 mM MgSO4, 0.4

immediately before use. In K+ stimulation studies, constant osmolality

D-TYR and/or (−) quinpirole hydrochloride (Tocris) were dissolved in
artificial cerebrospinal fluid (aCSF; 126 nM NaCl, 2.5 nM KCl, 1.2 nM NaH2PO4,
2.4 nM CaCl2, 1.2 nM MgCl2, 25 nM NaHCO3, 11 nM glucose, 0.4 nM l-ascorbic acid,
pH adjusted to 7.4) immediately before use.
