Human PC cell lines PANC-1, CFPAC-1, and Patu8988 (ATCC, Manassas, VA, USA) were grown in Dulbecco’s modified Eagle medium (DMEM; Invitrogen, Carlsbad, CA) that contained 10% fetal bovine serum and 1% penicillin–streptomycin (Sigma, Taufkirchen, Germany). Primary antibodies for NUDCD1 (Ab126902, 1:1000) and GAPDH (Ab8245, 1:2000) were obtained from Abcam. Antibodies against human Bcl-2 (12789-1-AP, 1:1000), Bax (50599-2-Ig, 1:1000) and Caspase 3 (19677-1-AP, 1:1000) were purchased from Proteintech. Antibodies against vimentin (AF7013, 1:1000), N-cadherin (AF4039, 1:1000), Cleaved-Caspase 3 (AF7022, 1:1000) and E-cadherin (AF0131, 1:1000) were purchased from Affinity Biosciences.

Investigating Protein Expression in Liver Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Total protein from L-02, SMMC7721, or HepG2 cells was extracted using a protein extraction kit (Solarbio, China). The protein concentration was quantified with the Bradford approach. Protein samples (50 µg) were separated via denaturing sodium dodecyl sulfate-polyacrylamide gel electrophoresis, followed by transference onto polyvinylidene fluoride membranes. Membranes were blocked by 5% skim milk then incubated with primary antibodies targeting NUDCD1 (1/5,000; ab126902; Abcam, USA), GAPDH (1/5,000; ab199553), GPX4 (1/2,000; ab252833), and ferritin heavy chain 1 (FTH1) (1/2,000; ab75972), followed by a secondary antibody. Protein bands were developed with an enhanced chemiluminescence reagent.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
