For cell-surface staining, aliquots of 106 splenic mononuclear cells were incubated with fluorescein isothiocyanate (FITC)-conjugated anti-mouse CD4 (GK1.5) antibody (eBioscience, San Diego, CA, USA). For intracellular staining, cells stained with CD4 were incubated with fixation/permeabilization working solution, and Fc receptors were blocked with excess mouse Fc block.
