Cell Proliferation Analysis via EdU Staining

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cell-Light EdU Apollo567 In Vitro Kit (100T) (C10310-1) was provided by Guangzhou RiboBio Co., Ltd. (Guangzhou, Guangdong, China). This kit was performed to detect cell proliferation. Firstly, HUVECs (4x103 cells per well) were seeded onto 96-well plates. After that, the cells were cultured with 50 μM EdU for 2 h followed by washed using PBS. Then, cells were incubated with 100 μL PBS containing 4% paraformaldehyde for 30 min. After that, cells were incubated with 100 μL 1 mg/mL DAPI for 10 min at room temperature in the darkness. Finally, a fluorescence microscope was used to observe the results of staining.

Assessing HUVEC Proliferation with EDU

Check if the same lab product or an alternative is used in the 5 most similar protocols
The biocompatibility of PEP@GO and the impact of the above experimental treatments on HUVEC cell proliferation were assessed using EDU uptake assays. HUVECs were placed in 24-well plates and treated with appropriate sEVs or miRNA formulations for 24 h. Following that, a Cell-Light EDU Apollo567 In vitro Kit (100 T) (RiboBio Co., Ltd., China) was utilized according to the instructions.

C2C12 Cell Proliferation Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
Near-confluent C2C12 cells (80–90%) were transfected with a vector overexpressing 2310043L19Rik (lnc-231), a control vector (pcDNA3.1), silnc-231, or NC silnc-231 (double strand). And the co-transfection group, transfected with pcDNA3.1+ NC mimic, miR-125a-5p, lnc-231+ miR-125a-5p, and lnc-231, respectively. After 12 h of transfection, C2C12 cells were cultured in fresh GM that contained 5-ethynyl-29-deoxyuridine (EdU; final concentration, 10 mM) for 4 h. An EdU assay with the Cell-Light EdU Apollo567 In Vitro Kit 100T (RIBOBIO, Guangzhou, China) was performed to test the proliferation of C2C12 cells and analyzed by confocal microscopy (LSM 700 META; Zeiss, Jena, Germany) to detect the EdU stained cells.

EdU Cell Proliferation Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
The cell proliferation was also evaluated using the Cell-Light EdU Apollo567 In Vitro Kit (100T) (RiboBio, Guangzhou, China). The cultured medium was aspirated after 48 h of transfection and incubated with EdU medium for 6 h, and the EdU-positive cells were detected following the manufacturer’s protocol.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
