Genetic Mouse Models for Neuroscience

The following mouse strains/lines were used in this study: C57BL/6J (The Jackson Laboratory, Stock # 000664), Pet1-Cre (The Jackson Laboratory, Stock # 012712), Pet1-Flpe (from the lab of Susan Dymecki [Jensen et al., 2008 (link)]), Pdyn-IRES-Cre (The Jackson Laboratory, Stock # 027958), and R26-LSL-FSF-ReaChR:mCitrine (The Jackson Laboratory, Stock # 024846). Animals were kept on a 12:12 regular light/dark cycle under standard housing conditions. All procedures were performed in accordance with protocols approved by the Harvard Standing Committee on Animal Care following guidelines described in the U.S. National Institutes of Health Guide for the Care and Use of Laboratory Animals.

Mouse Genetic Tools for Neuroscience

All animal procedures followed the ethical guidelines outlined in the NIH Guide for the Care and Use of Laboratory Animals, and all protocols were approved by the Institutional Animal Care and Use Committee at Harvard Medical School. Animals were maintained under constant temperature (23 ± 1 °C) and relative humidity (46 ± 5%) with a 12-h light–dark cycle. Mice used in this study were from both sexes and of a mixed genetic background. Glp1r-ires-cre (029283), Gpr65-ires-cre (029282) and Crhr2-ires-cre (033728) mice were previously made in the laboratory and are now available at the Jackson Laboratory. Slc17a6-ires-cre (Vglut2-ires-cre, 028863), Slc32a1-ires-cre (Vgat-ires-cre, 028862), Rosa26-lsl-Gfp-L10a (024750), Tac1-ires-cre (021877), Pdyn-ires-cre (027958), Penk-ires-cre (025112), Cartpt-ires-cre (028533), Sst-ires-cre (013044), Th-cre (021877) and Gcg-cre (030542) mice were purchased from the Jackson Laboratory.

Gabra1-IRES-cre, lsl-L10-GFP and Ptger3flox mice were previously generated14 (link),22 (link),51 (link).

Transgenic Mouse Husbandry and Care

In these experiments, we used adult male and female C57BL/6J mice from Jackson Labs (Bar Harbor, ME, USA), as well as transgenic Sert-cre (Jackson Labs, 014554), Pdyn-ires-cre (Jackson Labs, 027958), and EGFP-L10a mice (Jackson Labs, 024750) which were bred in-house. Tph2-p2A-flpO mice were developed at the University of Iowa Genome Editing Core Facility. All mice were maintained at a temperature- and humidity-controlled AAALAC approved vivarium at the University of Iowa College of Medicine with ad libitum access to food and water and 24 hour on-call veterinary care.
