Murine Spleen B Cell Immunophenotyping

Single-cell suspensions from murine spleens (106 cell per test) were stained with fluorochrome-conjugated monoclonal antibodies (mABs) in a single tube containing anti-mouse CD19-Alexa Fluor 647 (BioLegend 115522), IgD-Alexa Fluor 488 (BioLedend, 405718), IgM-Brilliant Violet 421 (BioLegend, 406517) and CD27-PE (BioLegend 124209) antibodies (1:20 dilution) for 30 min at 4°C. Cell-associated fluorescence was assessed with an LSR-II flow cytometer and analyzed using FACSDiva software (both BD). Two independent experiments were conducted, comparing the percentage of CD19+ cells (B cells) from splenocytes between inducible Clec16a knock down mice after tamoxifen treatment and the littermates treated with oil as a control. Data were combined together and B cell percentage was compared via two-sided T-test.
