Preparation and Dissolution of Dioscin for In Vivo and In Vitro Studies

Dioscin was obtained from Sigma‐Aldrich (SMB00576, USA). We fully dissolved Dioscin in 0.5% sodium carboxymethyl cellulose (CMC‐Na, IS9000, Solarbio) solution in distilled water for in vivo experiments, and we fully dissolved Dioscin in 0.01% dimethyl sulfoxide (DMSO, D8371, Solarbio) for in vitro experiments.23

Antifungal Activity Evaluation of Dioscin

C. albicans SC5314, C. albicans ATCC10231, Candida glabrata ATCC2001, and Candida parapsilosis ATCC22019 were obtained from China General Microbiological Culture Collection Center (CGMCC). All these strains were maintained on YPD (yeast extract 1%, peptone 2%, dextrose 2%) medium and stored at -80°C. Before each test, a colony from fungal strains subcultured twice on YPD agar was transferred into YPD medium and incubated overnight at 30°C with a speed of 140 rpm for propagation.
Dioscin was bought from Solarbio Life Sciences. N-acetyl-D-glucosamine (GlcNAc), XTT, MTT, menadione, propidium iodide (PI), and RPMI-1640 medium were bought from Sigma.

Quantitative Analysis of Dioscin in M. charantia

The standard of dioscin and 1-butly-3-methylimidazolium tetrafluoroborate ([C4MIM]BF4, ≥97.0%, CAS: 174501-65-6) were purchased from Solarbio (Beijing, China). Vanillin (≥99.0%, CAS: 121-33-5) was obtained from Tianjin Kemiou Chemical Reagent Co., Ltd. (Tianjin, China). Chromatographic grade methanoic acid (≥98.0%, CAS: 64-18-6) and acetonitrile (≥99.9%, CAS: 75-05-8) were obtained from Sigma-Aldrich-Fluka (Shanghai, China) and used for high performance liquid chromatography (HPLC) analysis. Other kinds of solvent used in the experiment, such as ethanol, were analytical grade and purchased from Sichuan Xilong Chemical Works (Chengdu, China). M. charantia powder (M. charantia pericarp and inner tissue were dehydrated, then vacuum dried) was purchased from Hunan Changsha Bojian Biological Technology Co., Ltd. (Changsha, China).

Dioscin-induced Apoptosis Regulation in Cells

Dioscin (purity ≥98%) was purchased from Beijing Solarbio Science & Technology Co., Ltd. Cells were treated with different concentrations of Dioscin (0, 0.5, 1 or 2 µM) for 24 or 48 h, or with 1 µM Dioscin for different durations (0, 2, 4 or 8 h). Nuclear Protein Extraction kit (cat. no. R0050) was purchased from Beijing Solarbio Science & Technology Co., Ltd and was used to extract nuclear proteins prior to western blotting. Primary antibodies targeted against GAPDH (cat. no. 60004-1-Ig) and histone H3 (cat. no. 17168-1-AP) were obtained from ProteinTech Group, Inc. Primary antibodies targeted against LATS1 (cat. no. 3477), p-LATS1 (Ser909; cat. no. 9157), p-MST (cat. no. 49332), MST2 (cat. no. 3952), YAP (cat. no. 4912), p-YAP (Ser397; cat. no. 13619), cyclin D1 (cat. no. 92G2) and BAX (cat. no. 5023) were purchased from Cell Signaling Technologies, Inc. The primary antibody targeted against BCL-2 (cat. no. WL01556) was purchased from Wanleibio Co., Ltd. The primary antibody targeted against RASSF1A (cat. no. ab91212) was purchased from Abcam. The goat anti-mouse or anti-rabbit HRP-conjugated secondary antibody (cat. nos. S0002 or S0001) was obtained from Affinity Biosciences.
