Total RNA Isolation and cDNA Synthesis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Total RNA was isolated from 0.02 g of tissue using the Ribopure Kit (Ambion, UK) according to the manufacturers’ instructions. Contaminating DNA was removed by On-column PureLink® DNase I treatment (Ambion). The quantity, quality and integrity of the RNA samples were determined using a NanoDrop ND-1000 spectrophotometer (Labtech International Ltd.) and Agilent 2200 TapeStation system (Agilent Technologies); all had an RNA Integrity Number of >7.3. cDNA synthesis from 1 μg RNA was by SuperScript™ II Reverse Transcription Kit (Invitrogen, UK) using oligo-dT(15) primer and RNaseOUT (Invitrogen).

Isolation of High-Quality RNA from Abomasal Lymph Nodes

Check if the same lab product or an alternative is used in the 5 most similar protocols
Abomasal (gastric) lymph nodes (ALN) were removed immediately post mortem and stored at –80˚C in RNAlater (Ambion, UK). Total RNA was isolated using the Ribopure Kit (Ambion) according to the manufacturers’ instructions. Contaminating DNA was removed by On-column PureLink® DNase I treatment (Ambion). The quantity, quality and integrity of the RNA samples were determined using a NanoDrop ND-1000 spectrophotometer and Agilent 2200 TapeStation system; all had an RNA Integrity Number of > 7.5.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
