Ethylnitrosourea-Induced Mutagenesis in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Male C57BL/6J mice (CLEA Japan) were treated with ethylnitrosourea (85 mg/kg, Sigma-Aldrich) by intraperitoneal injection twice at weekly intervals at the age of 8 weeks. At the age of 25-30 weeks, the sperm of the mice was used for in vitro fertilization with eggs of C57BL/6N mice to obtain F1 offspring. Mice were provided food and water ad libitum, and were maintained on a 12-hour light/dark cycle and housed under controlled temperature and humidity conditions. All procedures were approved by the Institutional Animal Care and Use Committee of the University of Tsukuba and the RIKEN BioResource Center, University of Texas Southwestern Medical Center at Dallas.

Male C57BL/6J mice (CLEA Japan) were treated with ethylnitrosourea (85 mg/kg BW, Sigma-Aldrich) via intraperitoneal injections twice at weekly intervals at 8 weeks old17 (link). At the age of 25–30 weeks, the sperm of the mice were collected and used for the in vitro fertilization of eggs from C57BL/6N mice to obtain F1 offspring. Mice were provided food and water ad libitum, maintained on a 12-hour light/dark cycle and housed under controlled temperature and humidity conditions. All procedures were carried out in accordance with the Guidelines for Animal Experiments of the University of Tsukuba and the RIKEN Tsukuba Institute and were approved by the Institutional Animal Care and Use Committee of the University of Tsukuba and the RIKEN Tsukuba Institute.

NDMA Mutagenicity Assay Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
N-Nitrosodimethylamine (NDMA, CAS no. 62-75-9) purity 95% (Batch Number, 2020-0369507) was purchased from Enamine Ltd (Kyiv, Ukraine) and was stored at approximately 4°C protected from light. Dose formulations were prepared in 0.5% (w/v) aqueous methylcellulose (400 cps at 2%) and were stable and homogenous for 8 days when protected from light and stored at 15–25°C or for 16 days at 2–8°C. Fresh formulations were prepared weekly for the main Muta™Mouse study. Ethyl nitrosourea, cyclophosphamide, and ethyl methane sulphonate were purchased from Sigma (UK).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
