Nicotine tartrate salt

(−)Nicotine tartrate salt (MP Biomedicals, Solon, OH) was dissolved in 0.9% sterile saline and adjusted to pH 7.2–7.4 with 1 M NaOH. The final stock concentration was 0.2 mg/mL free base, which was diluted to 0.02 mg/kg/mL based on body weight. DREADD Viral vectors AAV5-hsyn-DIO-mcherry (Catalog # 50459) and AAV5-hsyn-DIO-HM4D(Gi)-mcherry (Catalog # 44362) were obtained from Addgene (Watertown, MA, USA). The AAV5-hsyn-DIO-rM3D(G_s)-mcherry plasmid (Catalog # 50458) was obtained from Addgene and packaged using Penn Vector Core (Philadelphia, PA, USA). Heparin was purchased from Sagent Pharmaceuticals (Schaumburg, IL). Xylazine (100 mg/mL) was purchased from Biomeda (Cambridge, ON, Canada) and diluted to 8 mg/kg/mL with sterile saline prior to use. Ketamine was purchased from Dechra Veterinary Products (Overland Park, KS). Meloxicam and Cefazolin were both purchased through Henry Schein (Melville, NY) and diluted to 1 mg/kg and 100 mg/kg in sterile saline, respectively.

All nicotine doses utilized were based on free base weight and expressed as mg/kg/infusion. Ketamine (Akorn Animal Health, Lake Forest, IL), xylazine (Akorn Animal Health, Lake Forest, IL), cefazolin (Qilu Pharmaceutical, Shandong, China), meloxicam (Norbrook Inc., Overbrook, KS), and heparin (Sagent Pharmaceuticals, Schaumburg, IL) were administered as discussed above.
