Fentanyl and 2H5-Fentanyl were purchased from Cerilliant (Round Rock, TX). Carfentanil, sufentanil, and their corresponding N-phenyl-2H5 labeled forms, as well as 13C6-alfentanil HCl were custom synthesized by Battelle (Columbus, OH). Lofentanil, alfentanil, 3-methyl Fentanyl, and α-methyl Fentanyl, were generous gifts from a variety of sources listed in the Acknowledgements. Structures of all target analytes are shown in Figure 1. High-performance liquid chromatography (HPLC)-grade methanol and acetonitrile were purchased from Tedia Company, Inc. (Fairfield, OH). Formic acid (99%) and ammonium hydroxide (28.6%) were purchased from Sigma Aldrich (Pittsburgh, PA). Deionized (DI) water (>18MΩ·cm) was prepared on-site using an installed water purification system (Aqua Solutions, Inc., Jasper, GA). Pooled whole blood and a convenience set of individual whole blood samples were purchased from Tennessee Blood Services (Memphis, TN). Use of deidentified spiked blood from a commercial source was not deemed to constitute human subject research.

Formulation and Evaluation of Warfarin Capsules

LAA was obtained from Tianjin Guangfu Fine Chemical Research Institute (Tianjin, People’s Republic of China). HPMC (3.23 mPa.s), F127, and hard gelatin capsules (clear, size 00) were purchased by Sigma-Aldrich (St Louis, MO, USA). Warfarin was provided by the Central Research Institute, Shanghai Pharmaceuticals Holding Co, Ltd (Shanghai, People’s Republic of China). High-performance liquid chromatography (HPLC)-grade methanol was purchased from Tedia (Fairfield, OH, USA). CO2 (purity: 99.9%) was obtained from Huaming Gas Sales Co, Ltd (Tianjin, People’s Republic of China). Dichloromethane (analytical grade) was purchased from Sinopharm Chemical Reagent Co, Ltd (Shanghai, People’s Republic of China). Deionized water was prepared using a Milli-Q purification system (Millipore, Billerica, MA, USA). All other chemicals were of analytical grade and used as received.

Ethyl oleate, oleic acid, oleoyl macrogolglycerides (Labrafil M 1944 CS), and medium-chain triglycerides were obtained from Gattefossé (France). Polyoxyethylene stearate (Solutol) and polyethylene glycol 400 (PEG400) were purchased from Sinopharm Chemical Reagent (Shanghai, China). Cabomer-940 was purchased from Shin-Etsu Chemical Japan. Lactate chitosan (LC), carboxymethyl chitosan (CC), acetic chitosan (AC), and hydroxypropyl chitosan (HC) were purchased from Sinopharm Chemical Reagent (Shanghai, China). 4,8,5′-Trimethylpsoralen (TMP) was purchased from Tauto Biotech (Shanghai, China). High-performance liquid chromatography (HPLC)-grade methanol and acetonitrile were obtained from Tedia (Fairfield, OH, USA). Double-distilled water was used throughout the study. All other chemical reagents and solvents were of analytical grade.

HPLC-MS/MS Quantification of HIV Drugs

Standards, 3TC, AZT, NVP, EFV, RTV, and LPV were purchased from TargetMol (Shanghai, China); corresponding deuterated internal standards (IS), 3TC-d3, AZT-d4, NVP-d3, EFV-d5, RTV-d6, and LPV-d8 were obtained from Toronto Research Chemicals (Toronto, ON, Canada). Ammonium acetate and high-performance liquid chromatography (HPLC)-grade methanol were supplied by Tedia (Fairfield, OH, USA) and Sigma Aldrich (St. Louis, MO, USA), respectively. Tri-distilled water was from Watsons (Hong Kong, China).
Stock solutions of 3TC, NVP, and their corresponding ISs were individually dissolved in methanol at a concentration of 1 mg/mL. AZT, EFV, RTV, and LPV were used at a concentration of 400 μg/mL, with 50 μg/mL IS concentrations. They were all stored at −20°C. The working solutions with mixed six drug standards were obtained by diluting stock solutions with methanol to final concentrations at 0.1–10000 ng/mL. IS solutions were diluted with methanol at 5 ng/mL for 3TC-d3, NVP-d3, RTV-d6 and LPV-d8, 20 ng/mL for AZT-d4, and 100 ng/mL for EFV-d5.
