Extraction and Purification of Pristimerin

It is a triterpenoid quinone methide, and one of the bioactive components of the extracts prepared from the Celastraceae family of plants [25 (link)–28 (link)]. Purified Pristimerin (C30H40O4) was purchased from Pi and Pi Technology, Guangzhou, China. A stock solution of Pristimerin, 10 mg in 0.6 ml of dimethylsulfoxide (DMSO) (Sigma-Aldrich) was prepared and frozen at −20 °C in small aliquots until needed. PBS containing 1.2% DMSO was used both for dilution of the stock Pristimerin and as a ‘vehicle’ control for Pristimerin in different experiments. (For simplicity, PBS-DMSO has been denoted as PBS.) The dose of Pristimerin (1 mg/kg) used in vivo in this study is based on our titrations in a pilot study. The dose of Pristimerin used for in vitro testing was much below the dose that was cytotoxic for M3CT3 and lymph node cells (LNC) (data not shown).

Pristimerin-Induced Apoptosis in Cancer Cells

Pristimerin was purchased from Sigma Chemicals (St. Louis, MO). Anti-caspase-3 and caspase-9 antibodies were purchased from BD Pharmingen (San Diego, CA). Anti-PARP-1, anti-Bcl-2, anti-Bcl-xL and anti-survivin antibodies were purchased from Santa Cruz Biotechnology (Santa Cruz, CA). 96 AQueous One Solution Proliferation Assay System was from Promega (Madison, WI). Annexin V-FITC apoptosis detection kit was purchased from BD Pharmingen and mitochondrial potential sensor JC-1 was obtained from Molecular Probes, Invitrogen (San Diego, CA). Caspase-3 activity kit was from Clontech Laboratories (Mountain View, CA). Stock solution of PM (100 mM) was prepared in DMSO and all test concentrations were prepared by diluting stock solution in tissue culture medium.
