Modular p800 automatic analyzer

In sera of all rats from the two experiments, intact FGF23 concentration was measured by ELISA (Kainos Laboratories, Tokyo, Japan) as described in Heijboer et al [29 (link)]. Serum inorganic phosphorus and calcium were analyzed using a Modular P800 automatic analyzer (Roche, Mannheim, Germany).

Plasma glucose was measured in the laboratory of Turku PET Centre in duplicate using the glucose oxidase technique (Analox GM7 or GM9, Analox Instruments Ltd., London, UK). Glycosylated hemoglobin (HbA_1c) was measured with ion-exchange high performance liquid chromatography (Variant II Haemoglobin A_1c, Bio-Rad Laboratories, CA, USA). Plasma insulin was determined by time-resolved immunofluorometric assay (AutoDELFIA, Perkin Elmer Inc, Wallac Oy, Turku, Finland). Serum FFA were measured with a photometric enzymatic assay (Wako Chemicals GmbH, Neuss, Germany) on Modular P800 automatic analyzer (Roche Diagnostics, Mannheim, Germany). Serum high-sensitivity C-reactive protein (hs-CRP) was analysed with the sandwich immunoassay method using an Innotrac Aio1 immunoanalyzer (Innotrac Diagnostics, Turku Finland). Serum adipokines were analysed in duplicate by using Milliplex Human Serum Adipokine (Panel A) kit [cat.no HADK1-61K-A] (MilliporeSigma, Burlington, MA, USA) containing interleukin-6 (IL-6), interleukin-8 (IL-8), tumor necrosis factor alpha (TNFα), and leptin.

Arterial blood samples (0.5 mL) were drawn from the femoral artery at baseline, R15, and R90. The same volume of balanced colloid solution (VOLUVEN, Fresenius Kabi, Bad Homburg, Germany), was infused to correct for the very mild transient hypovolemia. Blood gas values, hemoglobin concentration, hemoglobin oxygen saturation, and acid-base balance were determined directly after sampling (ABL 505 blood gas analyzer, Radiometer, Copenhagen, Denmark).
Plasma lactate levels were assayed in samples acquired at baseline and at R90 by an enzymatic colorimetric method (modular P800 automatic analyzer, Roche Diagnostics, Basel, Switzerland).
It should be noted that urine-based kidney function parameters (urine production, creatinine clearance rate / glomerular filtration rate, and renal sodium reabsorption [58 ]) could not be determined because of I/R-induced anuria in all groups.
