Adult male dissected brains were fixed in 4% paraformaldehyde (Electron Microscopy Sciences) for 25 minutes and labeled using a modification of protocols previously described36 (link). The following primary antibodies were used: anti-bruchpilot (mAb nc82, 1:30, Developmental Studies Hybridoma Bank)37 (link), monoclonal rabbit anti-GFP (1:200, Molecular Probes), and rabbit anti-Tdc2 (1;100, Covalab). Brains in Supplemental Fig. 4 were labeled with diluted primary and secondary antibodies from the same aliquot, imaged with the same settings, and processed without modifications with ImageJ. Immunohistochemical analysis of MeCP2 in adult male brains

Brain samples were mounted in a drop of Vectashield (Vector Laboratories Inc, Burlingame, CA) and Images were collected on an Olympus Fluoview FV1000 laser scanning confocal mounted on an inverted IX81 microscope and processed with Image-J 1.33 (NIH) and Adobe Photoshop (Adobe, CA).
