In Vivo Mouse Model of Chronic Kidney Disease

Eight to 12-week-old female C57BL/6-Ly5.2 and C57BL/6-Ly5.1 mice were obtained from Charles River Laboratories (Maastricht, The Netherlands). The animals were fed commercial rodent chow and acidified water ad libitum and were maintained in the animal facility of the Leiden University Medical Center under conventional conditions. In selected experiments, Cinacalcet was administered as an additive in rodent chow to CKD and sham controls from week 6 after CKD induction onwards. All mouse studies were approved and performed according to the guidelines of the relevant authorities of the Leiden University Medical Center/Leiden University the Netherlands and within the guidelines set by the Dutch government.

Animal Welfare Protocols for AML1/ETO Murine Models

AML1/ETO-Rosa26-eYFP-Rosa26-cre-ER Ly5.2, Rosa26-Cre-ER Ly5.2 and Ubi-Luc2 knock-in mice were bread locally (IEO and University of Milan). We used 8- to 12-week-old males and females (average weight - 20–25 g).

Conditional Knockout and Lineage Tracing of Cebpa in Hematopoietic Stem Cells

CebpaloxP/loxP mice (kind gift of Daniel G. Tenen) were previously described (Zhang et al., 2004 (link)). HSC-SCL-Cre-ERT transgenic mice were generously provided by Joachim Göthert (Göthert et al., 2005 (link)). R26R-EYFP transgenic reporter mice were generously provided by Dennis Roop (Srinivas et al., 2001 (link)). All mice were backcrossed for at least five generations on a C57BL/6 background and bred to be heterozygous for R26R-EYFP and heterozygous for HSC-SCL-Cre-ERT, while each litter contained a mixture of Cebpa genotypes, such that Cebpa+/+ and CebpaΔ/Δ littermates were used in experiments. To induce recombination, mice were treated i.p. with 2.5 mg of tamoxifen (prepared at 10 mg/ml in corn oil) daily for 3 d and were used for experiments 7 d after the final dose of tamoxifen. Genotyping of mice and detection of recombined product were as previously described (Zhang et al., 2004 (link)). C57BL/6 (Ly5.2) and congenic (Ly5.1) mice were ordered from Charles River Laboratories. All procedures were performed in accordance with the University of Colorado Anschutz Medical Campus Institutional Animal Care and Use Committee–approved animal protocols.

Preclinical Mouse Models for Leukemia Research

CB17-SCID and C57BL/6-Ly5.1 mice of 6-8 weeks old were purchased from Charles River (France and Germany, respectively). For imaging and biodistribution experiments, we used both the immunodeficient THP-1 xenograft model and the immunocompetent syngeneic C1498 mouse model. For the THP-1 xenografts, CB17-SCID mice were inoculated subcutaneously in the right flank with five million THP-1 cells that were diluted 1:1 with Matrigel (Corning, New York, USA). Tumor inoculation was performed under 2.5% isoflurane anaesthesia and subcutaneous tumor growth was allowed to a size of 250 ± 50 mm³. For the syngeneic C1498 mouse model, C57BL/6-Ly5.1 mice were inoculated intravenously with one million lentivirally transduced AXLhigh C1498 cells under 2.5% isoflurane anaesthesia. C1498 mice were assessed daily for signs of leukemic disease (e.g., paralysis, weight loss). Prior to imaging, mice were anesthetized with 18.75 mg/kg of ketamine hydrochloride and 0.5 mg/kg of medetomidine hydrochloride. All animal experiments and housing conditions were approved by the Ethical Committee for Use of Laboratory Animals of the Vrije Universiteit Brussel and executed in accordance with the European guidelines for animal experimentation (License Agreement: LA 1230281, Files No.: 20-281-3, 21-281-2, 22-281-4, 23-281-1 and 23-281-4).

Transcriptomic Analysis of Murine Hematopoietic Stem Cells

Tissue extraction for transcriptome studies and the in vivo shRNA screen were performed in Cold Spring Harbor Laboratories (NY, USA). RNAseq libraries for transcriptomic assembly and differential expression were prepared from female C57BL/6 (6–12 weeks old) purchased from Charles River (Wilmington MA). For the in vivo shRNA screen, female B6.SJL-Ptprca/BoAiTac (also known as LY5.1, 8–12 weeks old) were purchased from Taconic (Hudson, NY). All these experiments were approved by the Cold Spring Harbor Animal Care and Use Committee.
Bone marrow transplantations of modified HSCs and subsequent analysis of peripheral blood from these mice were performed at the Cancer Research UK Cambridge Institute (Cambridge, UK). C57BL6J (6–12 weeks old) were purchased from Charles River (Kent, England). C57BL/6-LY5.1 females were purchased from Charles River (Kent, England) and used at 9–12 weeks old. These animal procedures were conducted in accordance with project and personal licenses issued under the United Kingdom Animals (Scientific Procedures) Act, 1986.

C57BL/6 Mouse Strain Protocols

Knockout Mouse Strains for Immune Studies

Dectin-1−/− and MyD88−/− mice were purchased from The Jackson Laboratory; TLR2−/− were provided by Shizuo Akira (Osaka University, Osaka, Japan). All knockout mice have a C57BL/6 background and were maintained at the animal production service facilities (SCSIE, University of Valencia). Wild-type C57BL/6 mice (Envigo) were used as controls; wild-type CD45.1-positive allotype mice (B6.SJL-Ptprca Pepcb/BoyCrl strain, also known as C57BL/6-Ly5.1 [Charles River Laboratories]) and the transgenic mice (B6.Cg-Tg[CAG-DsRed*MST]1Nagy/J strain, also known as DsRed.T3 [The Jackson Laboratory]) were used for in vivo transplantation assays and/or for the in vitro coculture assays. Experiments were conducted with 8- to 12-week-old mice (regardless of sex). Experiments were approved by the Committee on the Ethics of Animal Experiments of the University of Valencia (permit numbers 2017/VSC/PEA/00207 and 2019/VSC/PEA/0030) and performed according to Spanish law under Reales Decretos 1201/2005 and 53/2013. All efforts were made to minimize suffering.
