USA). Poly(vinyl alcohol) (PVA, Mw 31,000–50,000), poly(dl-lactide-co-glycolic) acid (PLGA, lactide:glycolide 50:50,
Mw 38,000–54,000), poly(ethylene glycol) diacrylate (Mn 750)
(PEG-DA), and 2-hydroxy-40-(2-hydroxyethoxy)-2-methylpropiophenone

Gelatin Methacryloyl (GelMA) Hydrogel Synthesis

A 10 % (w/v) gelatin solution was made by dissolving gelatin powder from porcine skin (G1890, Sigma Aldrich, USA) in 100 mL PBS at 50 °C for 1 h and then 8 mL of methacrylic anhydride (276685, Sigma Aldrich, USA) was added using a burette to the gelatin solution and stirred at 50 °C for 2 h before an additional 100 mL of PBS was added. Subsequently, the solution was dialyzed for 5 days at 40 °C using dialysis bag (888–11530, Spectrum Chemical Mfg. Corp., USA) to remove impurities. The purified solution was filtered by a vacuum filtration cup with 0.22 μm pores and frozen to −80 °C and subsequently lyophilized. The frozen GelMA was lyophilized for 3 days. The prepolymer solution of the GMM was prepared by dissolving 10% (w/v) lyophilized GelMA and 0.5% (w/v) photoinitiator (2-Hydroxy40-(2-hydroxyethoxy)-2-methylpropiophenone, 410896–10G, Sigma Aldrich, USA) in cell culture media.

Hydrogel Synthesis with Tunable Properties

Hydrogels were prepared using acrylamide (acrylamide, AAm, Sigma) and methylenebisacrylamide (N,N′-methylenebisacrylamide, MBA, Sigma) as monomers. The total concentration of AAm was fixed at 20% (w/v) and prepared by adjusting the concentration of MBA between 0.02% to 0.3% (w/v). An initiator, Irgacure2959 (2-hydroxy-40-(2-hydroxyethoxy)-2-methylpropiophenone, Sigma), was added to reach a final concentration of 0.2% (w/v). The hydrogels were formed by exposure to ultraviolet (UV) light (365 nm, VILBER LOURMAT, 4W) for 10 min. Subsequently, the gels were punched into 8 mm diameter disks and incubated in deionized (DI) water until fully swollen before characterization. The resulting hydrogels demonstrated tunable properties, showcasing the versatility of this synthesis approach for creating hydrogels with a range of mechanical and swelling characteristics suitable for diverse applications.

Photo-crosslinked GelMA Hydrogel Formation

GelMA was prepared based on our previous report.[34 (link)] For making photo-cross-linked GelMA hydrogel, 10% (w/v) of GelMA and 0.5% (w/v) of 2-hydroxy-40-(2-hydroxyethoxy)-2-methylpropiophenone (410896, Sigma-Aldrich) as a photoinitiator (PI) were added in the corresponding cell culture media and dissolved at 70 °C for 3 min. On day 4, suspended BE-MLMs with mild pipetting were mixed with the GelMA dispersion with PI, and then the mixture was crosslinked under an ultraviolet (UV)-exposure at 20 mW/cm2 for 3 min.
