RNA Isolation and qPCR from ES Cell Organoids

Check if the same lab product or an alternative is used in the 5 most similar protocols
ES cells grown in organoids were collected from Matrigel droplets by dissociation with dispase, and the organoids were pelleted. PureLink RNA Micro Kit (Invitrogen) was used to isolate RNA according to the manufacturer’s protocol. Total RNA (200–300 ng) was used for cDNA synthesis using SuperScript IV Reverse Transcriptase (Thermo Fisher Scientific). qPCR was performed using 2× Power SYBR green reagents on the QuantStudio 7 Thermocycler (Life Technologies). A list of primers and genes is provided in Supplementary Table 3.

RNA Isolation and qPCR Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
The RNeasy Micro Kit (QIAGEN) was used to isolate RNA according to the manufacturer’s protocol. Determination of RNA concentration and purity was performed by OD measurement (ratio of OD at 260 nm to OD at 280 was >1.7) using a NanoDrop spectrophotometer (Thermo Fisher Scientific). cDNA was synthesized using the Verso cDNA synthesis kit (Thermo Fisher Scientific). qPCR was performed using Fast SYBR green reagents on the QuantStudio 7 Thermocycler (Life Technologies, Thermo Fisher Scientific).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
