Rearing and Tissue Collection for Transcriptomics of Mythimna separata

Check if the same lab product or an alternative is used in the 5 most similar protocols
The larvae of M. separata were reared on an artificial diet in the climate incubator and maintained under a photoperiod of 14:10 h (light:dark) at 27 °C and 75% relative humidity [58 ]. Adult moths were provided with 10 % (weight by volume) sucrose solution. Various tissues from fifth instar larvae, including heads, epidermis, fat bodies, Malpighian tubules, and guts, were immediately dissected, and hemocytes were collected by cutting the thoracic legs. Various tissues from three day-old unmated female or male moths including brains, thoraxes, abdomens, antennae, labial palps, wings, and legs were collected, and all samples were used for RNA extraction. The reagents used in the pharmacological ligands, including serotonin hydrochloride (5-HT), (±)-octopamine hydrochloride (OA), dopamine hydrochloride (DA), tyramine hydrochloride (TA), (±)-8-hydroxy-2-(dipropylamino) tetralin hydrobromide (8-OH-DPAT), forskolin, 5-methoxytryptamine (5-MT), methiothepin mesylate, α-methylserotonin maleate (αm-5-HT), WAY-100635, SB-258719, and SB-269970 hydrochloride were purchased from Sigma-Aldrich (St Louis, MO, USA). Ketanserin was purchased from MedChemExpress (Monmouth Junction, NJ, USA).

Pharmacological Validation of 5-HT7 Receptor

Check if the same lab product or an alternative is used in the 5 most similar protocols
Norepinephrine (NE) hydrochloride was purchased from Sigma Chemical Co (catalog # A7256, St. Louis, MO, USA; water). Endothelin-1 (ET-1) was purchased from Echelon Biosciences (catalog # 331–25; Salt Lake City UT, USA; water) and 5-carboxamidotryptamine (5-CT) from Tocris Chemicals (catalog # 0458; Bio-Techne, Minneapolis, MN, USA; water). The following compounds were used as putative inverse agonists at the 5-HT7 receptor, prepared as 10−2 M solutions (catalog #, company, solvent): SB269970 (catalog #120508; AbCam, Boston, MA, USA; water), methiothepin (catalog #M149; Sigma Chemical Co St. Louis MO USA; water), risperidone (catalog #HY-11018; Med Chem Express, Monmouth Junction, NJ, USA; dimethylsulfoxide, DMSO), ketanserin (catalog #HY-10562A; Med Chem Express, Monmouth Junction, NJ, USA; DMSO), and clozapine (catalog #C6305; Sigma Chemical Co, St. Louis, MO, USA; DMSO).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
