4800 maldi tof tof mass spectrometry

Manufactured by AB Sciex
Sourced in United States

The 4800 MALDI TOF/TOF mass spectrometry is a laboratory instrument used for the analysis of molecular species. It utilizes matrix-assisted laser desorption/ionization (MALDI) coupled with time-of-flight (TOF) mass spectrometry technology to detect and identify various compounds, such as proteins, peptides, and small molecules.

Purification and Characterization of Synthetic Peptides

Check if the same lab product or an alternative is used in the 5 most similar protocols
All chemicals and solvents were purchased from commercial suppliers and used without further purification unless stated otherwise. Preparative high pressure liquid chromatography (RP-HPLC) was performed on an Agilent 1260 Series system. Systems were run with 0–95% methanol/water gradient with a 0.1% TFA modifier. High-resolution Matrix-assisted laser desorption/ionization (MALDI) spectra were performed on a 4800 MALDI TOF/TOF mass spectrometry (Sciex) at the Mass Spectrometry and Purdue Proteomics Facility (PPF), Purdue University. Peptides were synthesized on a CEM Liberty Blue peptide synthesizer. Compounds were also characterized and confirmed by TLC-MS or MALDI-MS. The purity of final compounds was confirmed on a Waters LC-MS system and/or Agilent 1260 Series system. Systems were run with 0–40% methanol/water gradient with 0.1% TFA. The purity of all target compounds showed >95%.

The standard Fmoc-protected amino acids were purchased from CEM Corporation. Acetyl coenzyme A lithium salt was purchased from Sigma-Aldrich. ThioGlo4 was purchased from Berry & Associates. High-performance liquid chromatography (HPLC) grade solvents were purchased from Fisher Scientific. Matrix-assisted laser desorption/ionization (MALDI) spectra were acquired using positive-ion mode on a 4800 MALDI TOF/TOF mass spectrometry (Sciex). Peptides were synthesized on a CEM Liberty Blue peptide synthesizer. The purity of the peptides was confirmed by Agilent 1260 Series HPLC system, eluting with a 0–40% acetonitrile/water gradient with 0.1% (v/v) TFA. All the purity of final peptides showed >95%. The spectra of MALDI and HPLC are available in the Supporting information (Figure S3).

Spectroscopic Characterization of Purified Compounds

Check if the same lab product or an alternative is used in the 5 most similar protocols
All chemicals and solvents were purchased from commercial suppliers and used without further purification unless stated otherwise. 1H and 13C NMR spectra were carried out on Bruker Avance 500 MHz NMR spectrometer (Billerica, MA, USA) in deuterated solvents. MALDI spectra were performed on 4800 MALDI TOF/TOF mass spectrometry (Sciex, Framingham, MA, USA) at the Mass Spectrometry and Purdue Proteomics Facility (PPF), Purdue University. Peptides were synthesized by CEM Liberty Blue peptide synthesizer (Matthews, NC, USA). Crude products were purified by chromatography using silica gel, standard grade from DAVSIL® (code number 1000179164, 35–70 micron SC). Flash chromatography was performed on Teledyne ISCO CombiFlash Companion chromatography system (Chicago, IL, USA) on RediSep prepacked silica cartridges. Thin-layer chromatography (TLC) plates (20 cm × 20 cm) were purchased from Merck KGaA (Darmstadt, Germany). All the purity of target compounds showed >95%.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
