Dopamine Quantification via HPLC-ECD

Samples were thawed and injected into a system of ESA 584 pumps (ESA, Chelmsfor, MA) with a pre-column filter (Synergi Max-RP4u Security Guard, 150×4.6mm, Phenomenex Inc., Torrance CA, USA) and Max-RP cartridges (Phenomenex), as we performed previously (Masini et al., 2004 (link)). Mobile phase was delivered at 1mL/min and contained 100mM sodium phosphate monobasic (Fisher, Pittsburgh PA, USA), 0.1mM EDTA (Sigma), 0.25mM octanesulfonic acid (Sigma), and 5% acetonitrile (JT Baker). Samples and standards were injected at 20μL using ESA 542 at 4°C. Peaks were detected over 30 min using ESA CoulArray (−150 and 200 mV on the initial and final electrodes). Position and height of dopamine peaks were compared to standards (Sigma; diluted in aCSF). Standards were run in duplicate/12 samples. Dopamine detection limit was 13.7 nmol/mL. Peak chromatogram area was integrated and analyzed by CoulArray 3.05.

Regenerated cellulose dialysis membranes with a molecular weight cutoff of 3500 g/mol were obtained from Spectrum Laboratories.

Kinetics Studies Solution Preparation

Dopamine and 2-aminophenol were purchased from Alfa Aesar (Ward Hill, MA, USA). Sodium phosphate monobasic was received from Fisher Scientific (Waltham, MA, USA) and catechol was obtained from Acros (Geel, Belgium). All chemicals were used as received. The background electrolyte (10 mM phosphate buffer) was prepared weekly by dissolving the desired amount of solid NaH2PO4. The pH of the solutions was adjusted, when necessary, using either 1 mol·L−1 NaOH or 1 mol·L−1 HCl (Fisher Scientific) and measured using a glass electrode and a digital pHmeter (Orion 420A+, Thermo; Waltham, MA). Stock solutions of Dopamine, 2-aminophenol and catechol (10 mM each) were prepared by dissolving the desired amount of each compound in ultrapure water (≥ 18 MΩ·cm, NANOpure Diamond, Barnstead; Dubuque, Iowa). Working solutions were prepared by diluting the stock solutions in 10 mM phosphate buffer. Standard-grade PMMA plates (150 × 70 × 1.5 mm) were purchased from Gravograph (Duluth, GA, USA) and used to produce the microdevices herein described. Sylgard 184 silicone elastomer and curing agent were obtained from Dow Corning. Sugar, used for the formation of the PDMS sponge (decoupler), was food-grade and acquired in a local grocery store.

Preparation and Characterization of Chitosan-TPP Nanoparticles

CHCl was obtained as a kind gift from Medical Union Pharmaceuticals (Ismailia, Egypt). High-purity, low-molecular weight CS from crab shells in the form of powder, with a degree of deacetylation of 75–85%, and TPP were purchased from Sigma Aldrich (San Diego, CA, USA). Potassium chloride, sodium chloride, sodium phosphate monobasic and potassium phosphate monobasic were obtained from Fisher Scientific (Fair Lawn, NJ, USA), as the chemicals required to obtain phosphate buffer (PB) media for release studies. Methanol and acetonitrile (HPLC grades) and acetic acid (glacial) were acquired from BDH Laboratory Supplies (Poole, UK). Muller–Hinton broth was supplied by Oxoid (Basingstoke, UK). The bacterial strains P. aeruginosa ATCC® 27853 and S. aureus ATCC® 25923 were obtained from Pharmaceutical Microbiological Laboratory (King Saud University, Riyadh, Saudi Arabia). Purified water was used throughout, and it was obtained by means of a Milli-Q® water purifier (Millipore, Molsheim, France). All chemicals were of pharmaceutical and analytical grade and were used without further purification.
