Quantitative Western Blot Analysis of Cellular Signaling

Cells were harvested, fractionated and transferred onto nitrocellulose membranes as described previously (Kim et al., 2015 (link)). Antibodies to STING (D2P2F), phospho-TBK1 (D52C2) and LC3B (D11) were purchased from Cell Signaling Technology. An anti-FLAG M2 antibody (#200474) was purchased from Agilent technologies. Antibodies to tubulin (B-5-1-2) and TBK1 (AOW9) were purchased from Sigma–Aldrich and EMD Millipore, respectively. Anti-HCMV IE (CH160) antibody was purchased from Virusys. Antibodies to IE86 (12E2), c-Myc (9E10) and GST (B-14) were purchased from Santa Cruz Biotechnology. Secondary peroxidase-labeled anti-mouse or anti-rabbit immunoglobulin G antibodies were purchased from Jackson ImmunoResearch. The signal intensity of protein bands were quantitated using Image LabTM software (Bio-Rad Laboratories).

Western Blot Protein Analysis

Protein extracts were separated by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) on 7.5%–16% gradient gels and transferred by semi-dry blotting to Hybond ECL nitrocellulose membranes (GE Healthcare). The following primary antibodies were used to develop the blots: anti-MK2, anti-p38, anti-pT180/Y182-p38, anti-pT334-MK2, anti-p(S/T)-PKD-substrate, anti-Hsp27, anti-pS82-Hsp27 (all purchased from Cell Signaling), anti-GAPDH (Millipore), anti-eEF2, anti-GFP, anti-hRRP40 (all from Santa Cruz Biotechnology), anti-RBM7, anti-ZCCHC8 (both from Atlas antibodies), anti-pS136-Rbm7 (see above); anti-NOGO-B (Rousseau et al. 2005 (link)), and anti-CBP80 (Andersen et al. 2013 (link)) antibody were described before. The anti-FLAG (M2) antibody was purchased from Agilent. ECL detection was carried out on a chemo-luminescence detector (LAS 3000 FujiFilm).

Western Blot Analysis of TMSB4Y and Related Proteins

Whole-cell protein lysates were prepared using Laemmli sample buffer and resolved by SDS-PAGE using NuPAGE gels (Invitrogen), transferred to a 0.2 μm pore size Invitrolon polyvinylidene difluoride (PVDF) membranes (Invitrogen), and probed with primary antibody followed by incubation with horseradish peroxidase-conjugated secondary antibodies. Antibodies used include anti-TMSB4Y (clone 6G4) mouse monoclonal antibody (SAB1403013, Sigma Aldrich), anti-FLAG M2 antibody (200472-21; Agilent), anti-rabbit IgG HRP-linked antibody (7074, Cell Signaling Technology), anti-mouse IgG HRP-linked antibody (7076, Cell Signaling Technology), anti-GAPDH (D16H11) XP rabbit monoclonal antibody (5174, Cell Signaling Technology), and anti-β-actin (clone 13E5) rabbit monoclonal antibody (4970, Cell Signaling Technology).
