Ultra leaf purified anti human cd3

Manufactured by BioLegend
Sourced in United States

Ultra-Leaf purified anti-human CD3 is a lab equipment product that can be used to detect and quantify CD3-positive cells. It is a monoclonal antibody that specifically binds to the CD3 complex, which is expressed on the surface of T cells. The product is purified and can be used in various immunological applications, such as flow cytometry, immunohistochemistry, and cell activation studies.

T cells were isolated using Pan T magnetic isolation beads (Miltenyi Biotec) according to manufacturer protocol, and stained with 1 uM carboxyfluorescein succinimidyl ester (CSFE) in 1× PBS in 37°C 5% CO2 for 10 min before washing the samples with media. Stained T cells were co-cultured with autologous DCs at a ratio of 5:1 (T:DC) in DC media. 100 ng/mL of Ultra-Leaf purified anti-human CD3 (BioLegend 300413) was added to each sample, and anti-CD3/CD28/CD2, ImmunoCult (STEMCELL Tech) was used as a positive control. Samples were incubated for 72 hours at 37°C 5% CO2 before being harvested and analyzed using flow cytometry. T-cell proliferation was quantified as the division index and proliferation index as calculated by FlowJo V.10.8.1 software.
Division index: Total number of divisions/the number of cells at the start of the culture.
Proliferation index: Total number of divisions/cells that went into division.

CD4+ T-cell Proliferation Assay with Tetanus, CD3, and Proinsulin Epitopes

Check if the same lab product or an alternative is used in the 5 most similar protocols
Tetanus toxoid (AJ Vaccines, 10 LfU mL−1), ultra‐LEAF‐purified anti‐human CD3 (Biolegend, San Diego USA, Clone OKT3, 0.1 µg mL−1) and proinsulin (PI) or HIP epitopes (GL Biochem, Shanghai, China) were included in CD4+ T‐cell proliferation assays at a final concentration of 10 µM.31 Proinsulin epitopes included the following sequences: PI33‐63 (C‐peptide, Sequence: EAEDLQVGQVELGGGPGAGSLQPLALEGSLQ), PI48‐62 (PI48‐62, Sequence: PGAGSLQPLALEGSL) and PI40‐52 (PI40‐52, Sequence: GQVELGGGPGAGS). HIPs containing parts of the PI33‐63 sequence included the following: hEGGG:C‐peptide (Sequence: GQVELGGGEAEDLQV), hEL:A‐chain (Sequence: SLQPLALGIVEQCC) and hEGGG:IAPP2 (Sequence: GQVELGGGNAVEVLK).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
